摘　　要：以三区域单通道微流控纸芯片(μPADs)为分析平台,通过比色法对乙酰胆碱酯酶(AChE)抑制剂进行活性评价。在纸芯片Ⅰ区滴加酶解液,Ⅲ区加载显色剂,2种溶液在纸张毛细作用下相向流动,在Ⅱ区混合并发生显色反应,手机拍照记录结果,Photoshop软件分析色度值,显色强度与AChE活性抑制率成反比。实验测得盐酸多奈哌齐和黄连碱的IC_(50)值分别为33.8 nmol/L和1.69 mol/L,与紫外-可见分光光度法(UV)所得结果符合较好。μPADs在酶抑制剂活性评价中具有快速便捷、样品用量少、分析成本低等优点,可用于酶抑制剂类药物的筛选。The microfluidic paper-based analytical devices(μPADs)with three regions-single channel were proposed and were applied evaluate the activity of acetylcholinesterase(AChE)inhibitors by colorimetric detection.Enzymatic solution and chromogenic agent were dropped onto the zoneⅠandⅢof theμPADs,respectively.The two solutions flowed toward each other in the channel according to the capillary action of paper,and then mixed and reacted in the zoneⅡ.After that,the results were recorded with a mobile phone and analyzed using Photoshop software.The color intensity is inversely proportional to the activity of enzyme inhibitors.The method was used to detect IC 50 values of donepezil hydrochloride and berberine hydrochloride.The measured IC 50 values were 33.8 nmol/L and 1.69 mol/L,respectively,which were in good agreement with the results obtained by UV method.The method based onμPADs has advantages of fast and convenient analysis,small sample consumption,and low analysis cost.It has the potency to be applied for drugs of enzyme inhibitors screening.
