摘 要:在单因素实验基础上,通过正交实验优化了浸泡时间、煎煮时间、V(水)∶m(槐米)和煎煮次数对槐米总黄酮提取率的影响;以槐米配方颗粒的外观、溶化性、成型率和休止角作为评价指标,对稀释剂的种类及比例、干膏粉与稀释剂的配比、φ(润湿剂)、干燥时间和干燥温度进行考察,优化其成型工艺。结果表明,最佳提取工艺为浸泡时间60 min、煎煮时间60 min、煎煮2次、V(水)∶m(槐米)=30 mL/g。在该条件下,槐米总黄酮的提取率为7.72%。优化后的槐米配方颗粒成型工艺条件为以m(乳糖)∶m(糊精)=2∶1为稀释剂、槐米提取物干膏粉和稀释剂的质量比为1∶1、润湿剂为φ(乙醇)=80%溶液,60℃干燥0.5 h,颗粒成型较好,工艺稳定可靠。On the basis of single factor experiments,the effects of soaking time,decoction time,liquid to material ratio,and decoction times on the extraction rate of total flavonoids from Sophora japonica were optimized through orthogonal test.Using the appearance,solubility,molding rate,and angle of repose of Sophora japonica formula granules as evaluation indicators,the types and proportions of diluents,the ratio of dry extract powder to diluents,the concentration of wetting agents,drying time and temperature were investigated to optimize its molding process.The results showed that the optimal extraction process was soaking time of 60 minutes,decoction time of 60 minutes,decoction twice,and the ratio of water to the mass of Sophora japonica was 30 mL/g.Under these conditions,the extraction rate of total flavonoids from Sophora japonica was 7.72%.The optimal molding process for Sophora japonica formula granules was as follows:lactose and dextrin(2∶1)was used as diluent,the ratio of dry extract powder to diluent was 1∶1,ethanol solution(80%)was used as wetting agent,and dried at 60℃for 0.5 h.Under these conditions,the molding of Sophora japonica formula granules is effective,and the process is stable and reliable.
