摘  要:【目的】探究急性低氧胁迫对大口黑鲈TG代谢的影响。【方法】试验设置3个处理组[DO:(7.0±0.15)mg/L、(3.5±0.12)mg/L、(1.2±0.2)mg/L]和6个采样时间点(0、4、8、12、24 h和复氧12 h),测定急性低氧胁迫后大口黑鲈肝脏、心脏、脑和肠系膜4种组织中与TG(甘油三酯)代谢相关的生化指标[TG(甘油三酯)、FFA(游离脂肪酸)、LPS(脂肪酶)及LPL(脂蛋白脂肪酶)]酶活性,检测了相关基因[PPAR alpha(过氧化物酶体增殖物激活受体α)、PPAR beta(过氧化物酶体增殖物激活受体β)、PPAR gamma(过氧化物酶体增殖物激活受体γ)、LPL]的表达情况。【结果】急性低氧胁迫后,大口黑鲈4种组织中TG和FFA的含量显著升高(P<0.05),肝脏、心脏和脑中LPS酶活性显著上升(P<0.05)。在急性低氧胁迫8和12 h时,4种组织中PPAR alpha基因的表达量显著上调(P<0.05)。复氧后,肠系膜脂肪中的PPAR beta基因表达量恢复到正常水平。PPAR gamma基因在肝脏中表达量显著上调(P<0.05),在其他3种组织中表达量不明显。低氧还诱导了LPL基因出现差异性表达。【结论】急性低氧胁迫下大口黑鲈4种组织中FFA氧化被抑制,TG合成和水解速率发生改变。肝脏、心脏和脑3种组织中TG水解活跃,大口黑鲈很可能在厌氧条件下利用甘油通过3-磷酸甘油途径增加糖酵解通量优先保证这3种组织的能量供应。【Objective】This study aims to investigate the impact of acute hypoxia stress on triglyceride(TG)metabolism in largemouth bass.【Method】Three treatment groups[dissolved oxygen(DO):(7.0±0.15)mg/L,(3.5±0.12)mg/L,(1.2±0.2)mg/L]and six sampling time points(0,4,8,12,24 h and reoxygenation 12 h)were established in this experiment to measure biochemical markers related to TG metabolism,including TG,free fatty acid(FFA),lipase(LPS),and lipoprotein lipase(LPL)enzyme activity in liver,heart,brain,and mesenteric of largemouth bass following acute hypoxic stress.Additionally,the expression of genes related to lipid metabolism,including peroxisome proliferator-activated receptor alpha(PPARα),peroxisome proliferator-activated receptor beta(PPARβ),peroxisome proliferator-activated receptor gamma(PPARγ),and lipoprotein lipase(LPL),was analyzed.【Result】After acute hypoxic stress,the contents of TG and FFA in four tissues of largemouth bass were significantly increased(P<0.05),and the activities of LPS enzymes in liver,heart and brain were significantly increased(P<0.05).The expression of PPARαgene was significantly up-regulated(P<0.05)in the four tissues at 8 h and 12 h of acute hypoxic stress.After reoxygenation,PPARβgene expression in mesenteric fat returned to normal levels.PPARγgene expression was significantly up-regulated in liver(P<0.05)and insignificant in the other three tissues.Hypoxia also induced differential expression of the LPL gene.【Conclusion】Under acute hypoxic stress,the oxidation of FFAs is suppressed,and the rates of TG synthesis and hydrolysis were altered in four different tissues of largemouth bass.TG hydrolysis is active in liver,heart,and brain tissues,suggesting that largemouth bass may utilize glycerol through the 3-phosphoglycerate pathway to prioritize energy supply in these three tissues under anaerobic conditions.
