摘 要:【目的】四川某养殖场的患病金鱼肌肉隆起甚至皮肤破溃,严重影响其观赏价值,为明确病因开展了本研究。【方法】通过组织病理检查、寄生虫形态观察和PCR鉴定进行诊断。【结果】大体病理观察发现病鱼主要症状为背鳍前方肌肉隆起形成驼峰样病灶,病程后期病灶破裂形成溃疡;组织学检查发现背部肌肉组织的损伤最为严重,病灶内大量肌细胞变性坏死被寄生虫孢子所替代;形态学观察可见寄生虫壳面观呈椭圆形、缝面观呈梨形,大小为10.3~13.3μm×6.7~7.4μm,具有2个3.4~4.2μm×1.7~2.6μm的等大极囊,其内有4~5圈极丝垂直于极囊纵轴缠绕,寄生虫形态与碘泡虫形态基本一致;基于碘泡虫的18S rDNA扩增测序构建系统发育结果表明寄生虫为晶状缝碘泡虫。【结论】为金鱼晶状缝碘泡虫病的防治提供了基础资料。【Objective】This study was conducted to investigate the etiology of a disease affecting goldfish at a farm in Sichuan,which caused muscle bulging and even skin breakdown severely compromised their ornamental value.【Method】The diagnosis was made based on histopathological examination,morphological observation of parasites,and PCR identification.【Result】Gross pathological examination revealed that the primary symptom of the diseased fish was a hump-like lesion caused by the muscle bulge in front of the dorsal fin,which eventually ruptured and developed into an ulcer in the late stage of the disease.Histological analysis showed that the dorsal muscle tissue sustained the most severe damage,with numerous myoblasts in the lesion undergoing degeneration and necrosis,subsequently being replaced by parasite spores.Morphological observation indicated that the parasite had an oval shape on the shell surface and a pear shape on the suture surface,measuring 10.3-13.3μm×6.7-7.4μm.It possessed two equidistant polar vesicles measuring 3.4-4.2μm×1.7-2.6μm,with 4-5 rings of filaments perpendicularly entwined along the longitudinal axis of the vesicles,a morphology consistent with Myxobolus.Phylogenetic analysis of 18S rDNA amplification and sequencing confirmed that the parasite was Myxobolus lentisuturalis.【Conclusion】This study provides fundamental insights into the control of iodine vesicular disease in goldfish.
