摘  要:【目的】对小麦穗增长突变体isl1进行遗传分析、基因定位和表型效应评价。【方法】将isl1与K1和K4杂交,构建遗传分离群体;利用外显子捕获测序技术(BSE-seq),鉴定到一个控制穗长性状的关键遗传位点ISL1;开发连锁的分子标记,解析ISL1的遗传效应。【结果】将ISL1定位于5A染色体长臂上,最可能的候选基因为Q,该基因第一外显子的C/T错义变异可能导致了穗长突变表型的产生,并同时影响株高、穗粒数、小穗数和籽粒大小等性状。【结论】ISL1是Q基因的一种新的等位变异,对小麦株型具有多效性影响。【Objective】Genetic analysis,molecular mapping,and morphological effects of a wheat mutant with increased spike length(isl1)were investigated.【Method】Genetic segregation populations were constructed by crossing isl1 with the wild type K1 and a wheat variety K4.These population were used to identify a spike morphology regulatory gene,ISL1,using bulk segregant exome capture sequencing.A molecular marker tightly linked to ISL1 was developed and used to analyze the genetic effects of ISL1.【Result】ISL1 was mapped to chromosome 5A,and the most likely candidate gene is Q.A missense variation(C-T)in the first exon of Q may lead to the mutant phenotype of increased spike length,and pleiotropically affect plant height,grain number per spike,spikelet number per spike,and grain size,among others.【Conclusion】ISL1 may represent a novel allele of Q with pleiotropic effects on wheat shoot architecture.
