摘 要:【目的】通过表型分析和不同组织中的内源激素含量测定,明确分枝调控基因CmRAX2过表达对菊花‘神马’内源激素的影响,为调控菊花侧枝发育以及培育理想株型奠定基础。【方法】以单头切花菊‘神马’(Chrysanthemum morifolium'Jinba')为试验材料,以CmRAX2过表达株系381、382为研究对象,非转化菊花为对照,研究CmRAX2过表达对菊花分枝表型和内源激素含量的影响,讨论CmRAX2基因调控菊花分枝的机理。【结果】CmRAX2过表达促进菊花植株生长和侧芽的萌发,植株顶芽中吲哚乙酸含量增加、茉莉酸含量减少,根部吲哚乙酸和氨基环丙烷羧酸的含量增加,侧芽中吲哚乙酸和茉莉酸的含量降低、异戊烯基腺嘌呤核苷和氨基环丙烷羧酸的含量增加。【结论】CmRAX2过表达可通过影响吲哚乙酸、茉莉酸、氨基环丙烷羧酸和异戊烯基腺嘌呤核苷等内源激素含量调节植株生长、生根和侧芽发育。【Objective】Through phenotypic analysis and the determination of endogenous hormone content in various tissues,we clarified the effects of overexpression the branching regulatory gene CmRAX2 on the endogenous hormones of chrysanthemum'Jinba'.This research provides a foundation for manipulating the development of lateral branches and cultivating the desired plant type in chrysanthemums.【Method】We used single-head cut chrysanthemum'Jinba'(Chrysanthemum morifolium'Jinba')as the experimental material.CmRAX2 overexpression lines 381 and 382 were used as experimental groups,with wild-type chrysanthemums serving as the control.We studied the differences in branching phenotypes and endogenous hormone contents between the transformed and control chrysanthemums to explore the mechanism by which the CmRAX2 gene regulates chrysanthemum branching.【Result】Overexpression of CmRAX2 enhanced the growth of chrysanthemum plants and the germination of lateral buds.In the apical buds,the content of indoleacetic acid increased,while jasmonic acid decreased.In the roots,there was an increase in the content of indoleacetic acid and aminocyclopropanecarboxylic acid.In the lateral buds,the content of indoleacetic acid and jasmonic acid decreased,but the content of isopentenyl adenosine nucleotide and aminocyclopropanecarboxylic acid increased.【Conclusion】Overexpression of CmRAX2 can regulate plant growth,rooting and lateral shoot development by modulating the content of endogenous hormones such as indoleacetic acid,jasmonic acid,aminocyclopropanecarboxylic acid,and isopentenyladenine riboside.
