摘 要:为了分析湿加松Pinus elliottii×P.caribaea育种群体的遗传多样性和遗传结构,并构建核心种质,辅助湿加松高世代育种,研究以台山市红岭种子园内湿加松初级种子园的50个无性系和湿加松育种园的66个优良个体为材料,通过SLAF-seq技术开发126365个高质量SNP标记。遗传多样性分析结果显示,湿加松育种群体期望杂合度(He)、观测杂合度(Ho)分别为0.2806、0.2103,Nei多样性指数(H)、多态性信息含量(PIC)和香浓-维纳指数(I)分别为0.2819、0.2323和0.4419。系统发育树将所有样本分为3个大分支,群体结构最佳类群数量K=6,其中类群Q4包含的种子园样本最少。评选出了23份材料构建核心种质,遗传多样性覆盖度超过99%,分别来自6个类群,其中只有7个来自初级种子园。该研究表明湿加松育种群体具有中等的遗传多样性和多源的遗传结构,揭示了湿加松初级种子园相较于整个育种群体在遗传结构方面存在一定的缺陷。This study aims to analyze the genetic diversity and structure of Pinus elliottii×P.caribaea breeding population and to establish a core germplasm to assist in high-generation breeding.This study used 50 clones from the primary seed orchard and 66 superior individuals from the breeding orchard of P.elliottii×P.caribaea in Hongling Seed Orchard,Taishan city as materials.A total of 126365 high-quality SNP markers were developed through SLAF-seq technology.Genetic diversity analysis revealed that the expected heterozygosity(He)and observed heterozygosity(Ho)were 0.2806 and 0.2103,respectively.The Nei′s diversity index(H),polymorphism information content(PIC),and Shannon-Wiener index(I)were 0.2819,0.2323,and 0.4419,respectively.The phylogenetic tree divided all samples into three major branches.The optimal number of groups in the population structure was K=6,and group Q4 contained the fewest samples from the seed orchard.Twenty-three materials were selected to construct the core collection,with a genetic diversity coverage of over 99%.These materials were from six groups,and only seven of them were from the primary seed orchard.This study indicates that the P.elliottii×P.caribaea breeding population has moderate genetic diversity and a multi-source genetic structure,revealing certain deficiencies in the genetic structure of the primary seed orchard of P.elliottii×P.caribaea.
