山荆子烂皮病的病原菌鉴定与致病性验证

摘  要:为明确引起山荆子Malus baccata烂皮病的病原菌种类及其致病性,对哈尔滨市山荆子烂皮病进行病害调查、病斑组织采样、菌株分离纯化、病原菌形态学鉴定与系统发育分析,并向健康的山荆子离体水培枝条以烧伤、刺伤和无伤接种方法,验证病原菌致病性。结果表明:分离出的代表性菌株De-NEFU1与间座壳属菌株亲缘关系较近,为榆树间座壳菌Diaporthe eres;烧伤后接种处理组表现出典型的烂皮症状,刺伤后接种处理组也表现出较明显的溃疡症状。确定D.eres为引起山荆子烂皮病的致病菌,且山荆子为D.eres国内外首次报道的新寄主植物。To determine the pathogenic fungus and pathogenicity,an investigation into the stem canker disease of Malus baccata in Harbin including disease survey,sampling of infected tissue,isolation and purification of pathogen strains,morphological observation and systematic phylogenetic analysis of pathogenic fungus for identification were carried out.Hydroponically grown,healthy branches of M.baccata were inoculated using burn,puncture,and non-injury methods.The results of systematic phylogenetic analysis showed that the representative strain De-NEFU1 was closely related to Diaporthe,the strain De-NEFU1 was Diaporthe eres.The branches inocculated with D.eres after burnt treatment could produce typical stem canker symptoms,the same symptoms as those in field,the branches inocculated with D.eres after punctured treatment also showed obvious symptoms of stem canker.D.eres was the pathogen causing the stem canker of M.baccata,which was a new host for D.eres for the first report at home and aboard.
