摘 要:为探讨开放式有菌环境条件下玉玲花组织培养技术,解决实生播种繁殖率低的问题,以玉玲花幼嫩叶片、腋芽或顶芽的茎段为外植体在有菌条件下进行离体培养,探讨消毒时间、培养基、激素浓度和种类、初代培养影响因素,旨在建立离体繁殖技术。结果显示:有菌环境条件下玉玲花幼嫩叶片最佳消毒时间是3 min,带腋芽茎段最佳消毒时间是5 min;叶片愈伤组织诱导的最佳处理为MS+6-BA 1.0 mg·L^(-1)+2,4-D 0.5 mg·L^(-1)+蔗糖20 g·L^(-1)+琼脂4.5 g·L^(-1);愈伤组织增殖最佳激素配比是6-BA 2.5 mg·L^(-1)+NAA0.25 mg·L^(-1)+蔗糖20 g·L^(-1)+琼脂4.5 g·L^(-1);带腋芽或顶芽的茎段离体培养的最佳处理为MS+6-BA 1.5 mg·L^(-1)+NAA 0.5 mg·L^(-1)+蔗糖30 g·L^(-1)+琼脂5 g·L^(-1)。研究结果表明:有菌环境条件下可以实现玉玲花组织培养、获得愈伤组织和无菌苗。To explore the tissue culture technology of Styrax obassia Sieb.etZucc.under the open environment with bacteria,and to solve the problem of low reproduction rate of solid sowing.The stem segments of young leaves,axillary buds or apex buds of Yulinghua were cultured in vitro under the presence of bacteria,and the influencing factors of disinfection time,medium,hormone concentration and species,and primary culture were discussed in order to establish in vitro propagation technology.The best disinfection time of young leaves of S.obassia under bacterial environment is 3 min.The optimum disinfection time of the stem segment with axillary buds is 5 min.The best treatment for leaf callus induction was MS+6-BA 1.0 mg·L^(-1)+2,4-D 0.5 mg·L^(-1)+sucrose 20 g·L^(-1)+AGAR 4.5 g·L^(-1).The optimal hormone ratio of callus proliferation was 6-BA 2.5 mg·L^(-1)+NAA 0.25 mg·L^(-1)+sucrose 20 g·L^(-1)+AGAR 4.5 g·L^(-1).The optimal treatment for stem segments with axillary or terminal buds in vitro was MS+6-BA 1.5 mg·L^(-1)+NAA 0.5 mg·L^(-1)+sucrose 30 g·L^(-1)+AGAR 5 g·L^(-1).It is possible to obtain callus and bacteria-free vaccine by tissue culture under bacterial environment.
