摘 要:目的研究人鼻病毒(human rhinoviruses,HRV)感染对肥大细胞(human mast cell line 1,MHC-1)激活和免疫应答功能的影响,探讨HRV诱发过敏性哮喘急性发作或加重的免疫病理机制。方法将HMC-1细胞随机分为3组,除RV16病毒感染组外,紫外线灭活的RV16病毒和PBS分别作为阴性对照组和空白对照组。采用实时定量PCR检测HRV感染HMC-1细胞后病毒复制水平、肥大细胞激活相关分子FcεR1、CD203c、CD63的表达变化,ELISA方法检测肿瘤坏死因子α(tumor necrosis factor,TNF-α)、白细胞介素6(interleukin 6,IL-6)、细胞介素8(interleukin 8,IL-8)和白细胞介素10(interleukin 10,IL-10)的分泌量。结果HRV感染HMC-1细胞后,感染24 h病毒载量明显增加(P<0.05),感染48 h病毒载量达到最高(P<0.01);荧光定量PCR结果显示,与对照相比,RV16病毒感染后HMC-1细胞激活相关分子FcεR1、CD203c、CD63的表达量明显增加(P<0.01);ELISA检测结果显示,与对照组相比,RV16病毒感染后HMC-1细胞TNF-α、IL-6、IL-8和IL-10分泌量明显增加(P<0.05)。结论HRV感染促进肥大细胞激活和细胞因子分泌,与HRV引起的哮喘加重和发病机制密切相关。Objective To investigate the effect of human rhinoviruses(HRV)infection on the activation of human mast cell line 1(MHC-1)and the function of immune response,and to explore the immunopathological mechanism of acute attack or aggravation of allergic asthma induced by HRV.Methods HMC-1 cells were randomly divided into three groups.Except for the RV-16virus infection group,UV inactivated RV-16 virus and PBS were used as negative control group and blank control group,respectively.Virus replication in HMC-1 cells and the expression of FcεR1,CD203c,CD63 were quantified by real-time quantitative PCR(qRT-PCR).The levels of tumor necrosis factorα(TNF-α),interleukin 6(IL-6),interleukin 8(IL-8)and interleukin 10(IL-10)secreted by HMC-1cells were detected by ELISA.Results After HRV infected HMC-1 cells,the viral load increased significantly at 24 hours(P<0.05),and reached the highest at 48 hours(P<0.01);Fluorescence quantitative PCR results showed that compared with control,the expression levels of activation-related molecules FcεR1,CD203c and CD63 in HMC-1 cells were significantly increased after RV16 virus infection(P<0.01);ELISA results showed that compared with the control group,the secretion of TNF-α,IL-6,IL-8 and IL-10 in HMC-1 cells increased significantly after RV16 virus infection(P<0.05).Conclusion HRV promotes the mast cell activation and enhances the cytokines secretion from mast cells,which is closely related to the exacerbation and pathogenesis of asthma caused by HRV.
