摘 要:树突状细胞(Dendritic Cells,DC)作为抗原递呈细胞中的佼佼者,不仅具有强大的抗原递呈能力,还在免疫耐受中扮演着关键角色。如何利用DC的免疫耐受特性来治疗自身免疫性疾病,已成为免疫学研究的热点。重症肌无力作为一种T细胞依赖、抗体介导、器官特异的自身免疫病,其治疗也迎来了新的突破点——利用DC的免疫耐受特性。近年来,围绕DC的耐受特性、诱导耐受机制及其利用细胞因子诱导获得耐受性DC、DC负载乙酰胆碱受体或免疫优势肽段、基因修饰DC、特异性抑制核因子κB依赖的基因表达等治疗重症肌无力应用研究不断增多,为这一疾病的治疗提供了新的思路和方向。Dendritic cells(DC),as outstanding antigen-presenting cells,not only possess powerful antigen-presenting capabilities but also play a crucial role in immune tolerance.How to harness the immune tolerance characteristics of DC to treat autoimmune diseases has become a hot topic in immunological research.Myasthenia gravis,as a T cell-dependent,antibody-mediated,organ-specific autoimmune disease,has seen a new breakthrough in treatment—utilizing the immune tolerance properties of DC.In recent years,research on the tolerance characteristics of DC,the mechanisms of inducing tolerance,and the application of cytokine-induced tolerogenic DC,DC loaded with acetylcholine receptors or immunodominant peptides,genetically modified DC,and specific inhibition of nuclear factor-κB-dependent gene expression for the treatment of myasthenia gravis has been increasing.These studies provide new insights and directions for the treatment of this disease.
