梨星毛虫幼虫转录组及SSR位点分析

摘  要:为了探讨SSR位点分布特征,进而为开发、丰富新型EST-SSR分子标记奠定基础,采用Illumina HiSeq^(TM)2000高通量测序平台对梨星毛虫幼虫进行转录组测序,在此基础上筛选其微卫星位点并分析分布特征。结果显示,梨星毛虫幼虫转录组测序共获得52671条转录本,21432条unigenes,COG、GO注释和KEGG通路数据库中unigenes注释占主导地位的分别是一般功能预测、细胞进程、运输和分解代谢。8288个微卫星位点分布于5832条unigenes中,SSR重复类型主要为单核苷酸和二核苷酸重复,单核苷酸重复中最主要的基元类型是(A/T)n,占总SSR位点77.94%,(AC/GT)n为二核苷酸重复的优势基元,占总SSR位点数的2.38%,且每个母本(maternal blood)序列中SSR数目随着碱基重复类型的增加而减小,成反比出现。分析各类型的转录芯片数量占比,SSR二核苷酸重复类型的4个基序的转录芯片数量占比最大(48.03%),其中AT/AT的数量最大(228条)。研究结果实现了基于转录组数据成功筛选出梨星毛虫幼虫微卫星位点,为梨星毛虫的遗传多样性分析、基因挖掘等提供研究基础。In order to explore the distribution characteristics of SSR(simple sequence repeat)loci and thereby lay a foundation for the development and enrichment of novel EST-SSR-molecular markers,the larvae of the pear star moth were subjected to transcriptome sequencing using the Illumina HiSeq^(TM)2000 high-throughput sequencing platform.On this basis,microsatellite loci were screened and their distribution characteristics were analyzed.The results showed that a total of 52671 transcripts and 21432 unigenes were obtained from the transcriptome sequencing of Illiberis pruni.The dominant categories in the COG,GO annotations,and KEGG pathway database annotations of unigenes were general function prediction,cell process transport and catabolic.A total of 8288 microsatellite loci were distributed in 5832 unigenes.The main types of SSR repeats were mononucleotide and dinucleotide repeats.Among the mononucleotide repeats,the primary motif type was(A/T)n,accounting for 77.94%of the total SSR loci.The dinucleotide repeat(AC/GT)n was the dominant motif,representing 2.38%of the total SSR loci.The number of SSRs in each maternal blood sequence decreased with an increase in the base repeat types,showing an inverse relationship.In the analysis of the proportion of transcript chips from each type,the proportion of transcript chips of the four motifs of the SSR dinucleotide repeat types was the highest(48.03%),with the AT/AT motif being the most abundant(228 transcripts).The research findings facilitated the successful screening of microsatellite loci of Illiberis pruni based on transcriptome data,providing a research foundation for the genetic diversity analysis and gene mining of Illiberis pruni.
