通络益肾温阳方联合脐灸对糖尿病大鼠的影响

摘　　要：目的:探究通络益肾温阳方联合脐灸对糖尿病大鼠糖脂代谢、坐骨神经传导速度及基质金属蛋白酶(MMP)-3表达的影响。方法:选取50只SPF级SD雄性大鼠,随机分为正常组、模型组、通络益肾温阳方组、脐灸组、联合组,每组10只。对模型组、通络益肾温阳方组、脐灸组、联合组采用腹腔注射链脲佐菌素法建立糖尿病模型,建模成功后,通络益肾温阳方组给予通络益肾温阳方治疗,脐灸组给予脐灸治疗,联合组给予通络益肾温阳方联合脐灸治疗,正常组、模型组不予治疗。检测血清糖脂代谢指标;HE染色法检测肾组织病理形态;肌电采集系统检测坐骨神经传导速度;免疫组化法检测MMP-3表达相关蛋白。结果:与正常组相比,模型组空腹血糖(FBG)、空腹胰岛素(FINS)、胰岛素抵抗指数(HOMA-IR)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、MMP-3蛋白表达升高,而高密度脂蛋白胆固醇(HDL-C)、运动神经传导速度(MNCV)、感觉神经传导速度(SNCV)降低,差异有统计学意义(均P<0.05)。与模型组相比,通络益肾温阳方组、脐灸组、联合组FBG、FINS、HOMA-IR、TC、TG、LDL-C、MMP-3蛋白表达降低,而HDL-C、MNCV、SNCV升高,差异有统计学意义(均P<0.05)。联合组FBG、FINS、HOMA-IR、TC、TG、LDL-C、MMP-3、HDL-C、MNCV、SNCV与脐灸组比较,差异有统计学意义(均P<0.05)。结论:通络益肾温阳方联合脐灸能改善糖尿病大鼠糖脂代谢,提高坐骨神经传导速度,并对MMP-3表达具有调控作用。Objective:To explore effects of Tongluo Yishen Wenyang recipe combined with umbilical moxibustion on glucose and lipid metabolism,sciatic nerve conduction velocity and matrix metalloproteinase(MMP)-3 expression in diabetic rats.Methods:Fifty SPF SD male rats were selected and randomly divided into normal group,model group,Tongluo Yishen Wenyang formula group,umbilical moxibustion group and combined group,with 10 rats in each group.The model group,Tongluo Yishen Wenyang formula group,umbilical moxibustion group and combined group were modeled by intrabitoneal injection of streptozocin.Tongluo Yishen Wenyang formula group were treated with Tongluo Yishen Wenyang formula,umbilical moxibustion group treated with umbilical moxibustion,and the combined group treated with Tongluo Yishen Wenyang formula combined with umbilical moxibustion,while the normal group and model group were not treated.Serum glucose and lipid metabolism indexes were detected,pathological morphology of kidney tissue detected by HE staining method,sciatic nerve conduction velocity detected by electromyography collection system.The expression of MMP-3 expression related protein were detected by immunohistochemistry.Results:Compared with the normal group,fasting blood glucose(FBG),fasting insulin(FINS),insulin resistance index(HOMA-IR),total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C)and MMP-3 protein expression were increased in the model group.However,high density lipoprotein cholesterol(HDL-C),motor nerve conduction velocity(MNCV)and sensory nerve conduction velocity(SNCV)were decreased,difference statistically significant(all P<0.05).Compared with model group,the protein expressions of FBG,FINS,HOMA-IR,TC,TG,LDL-C and MMP-3 were decreased in Tongluo Yishen Wenyang formula group,umbilical moxibustion group and combined group,while HDL-C,MNCV and SNCV were increased,difference statistically significant(all P<0.05).The changes of FBG,FINS,HOMA-IR,TC,TG,LDL-C,MMP-3,HDL-C,MNCV and SNCV in combination group were higher t
