桃红四物汤对大鼠深静脉血栓防治机制研究

摘　　要：目的:观察桃红四物汤对深静脉血栓模型大鼠血栓重量、长度的影响及血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、血管细胞黏附分子-1(VCAM-1)、细胞间黏附分子-1(ICAM-1)的影响。方法:将32只雄性SD大鼠随机分为四组,桃红四物汤低、中、高组和模型组,每组8只。分别在造模术前使用不同剂量桃红四物汤连续灌胃,模型组大鼠灌胃等剂量蒸馏水。1周后四组均建立深静脉血栓模型,造模术后3 d各组大鼠脱颈处死,股动脉采血ELISA检测IL-6、TNF-α水平。截取下腔静脉及分支静脉结扎段,电子天平称取血栓重量,测量血栓长度。Western-blotting检测血栓VCAM-1、ICAM-1。比较各组上述指标。结果:术后,模型组对比,各组大鼠血栓长度、重量减小,差异有统计学意义(均P<0.01)。各组大鼠IL-6、TNF-α含量均下降(均P<0.01)。术后,各组大鼠ICAM-1、VCAM-1相对表达量均下降,组间差异有统计学意义(均P<0.01)。结论:桃红四物汤能减少血栓重量、长度,抑制血清IL-6、TNF-α炎症因子释放及血栓中ICAM-1、VCAM-1蛋白表达,对深静脉血栓形成有防治作用。Objective:To observe effects of Taohong Siwu decoction on weight and length of thrombus and effects of tumor necrosis factor-α(TNF-α)interleukin-6(IL-6)vascular cell adhesion molecule-1(VCAM-1)intercellular adhesion molecule-1(ICAM-1)in serum of rat model of deep vein thrombosis.Methods:32 male SD rats were randomly divided into four groups,Taohong Siwutang low,medium,high group and model group,with 8 rats in each group.The rats in the model group were given continuous intragastric administration of different doses of Taohong Siwu decoction and equal doses of distilled water before the modeling operation.Deep vein thrombosis model was established in four groups one week later,rats in each group were killed by neck removal 3 days after the modeling,levels of IL-6 and TNF-αwere detected by femoral artery ELISA.The ligation sections of inferior vena cava and branch vein were cut off.The weight and length of thrombus were measured by electronic balance.Thrombus VCAM-1 and ICAM-1 were detected by Western-blotting.Parallel inter-group comparison.Results:After operation,compared with model group,thrombus length and weight of rats in all groups decreased significantly,which was inversely proportional to dose of Chinese medicine,with statistical significance(P<0.01).IL-6,TNF-αcontents of rats in all groups decreased significantly,with statistical significance(all P<0.01).The relative expression levels of ICAM-1 and VCAM-1 were significantly decreased in all groups,difference statistically significant(all P<0.01).Conclusion:Taohong Siwu decoction can reduce weight and length of thrombus,inhibit the release of IL-6,TNF-αinflammatory factors in serum,and inhibit the expression of ICAM-1,VCAM-1 proteins in thrombus.Taohong Siwu decoction can prevent deep vein thrombosis.
