盐胁迫对不同水稻品种光合特性及基因表达的影响

摘  要:土壤盐碱化对水稻的生长和发育造成了严重的影响,探究盐胁迫对不同水稻品种光合作用及基因表达的影响,对盐碱地水稻高产和稳产具有重要意义。本研究以‘伊粳12号’和‘新稻36号’为试验材料,测定125 mmol∕L NaCl胁迫6 d的光合特性参数。结果表明,125 mmol∕L NaCl胁迫下‘新稻36号’最大荧光产量(Fm)、原初光能转化效率(Fv∕Fm)、净光合速率(Pn)、气孔导度(Gs)、胞间CO_(2)浓度(Ci)、蒸腾速率(Tr)和叶绿素相对含量值(SPAD)降低的幅度显著大于耐盐品种‘伊粳12号’。通过qRT-PCR分析发现,OsABCI7基因和OsHCF222基因在短时间内基因表达量均显著上调。亚细胞定位分析发现,OsABCI7基因编码蛋白定位于叶绿体,OsHCF222基因编码蛋白定位于细胞质。综上,‘伊粳12号’的光合系统防卫能力优于‘新稻36号’,该结果进一步为耐盐水稻资源筛选和鉴定提供理论依据。Soil salinization has serious effects on the growth and development of rice.It is important to investigate the effects of salt stress on photosynthesis and gene expression of different rice cultivacs for high and stable production of rice.The present study determined the photosynthetic characteristics of rice under the stress condition of 125 mmol∕L NaCl for 6 d. The experimental materials wer‘e yijing12’ and‘ xindao36’. The results showed that the decrease in maximum fluorescence yield( Fm)、primary light energy conversion efficiency (Fv ∕ Fm)、net photosynthetic rate (Pn)、stomatal conductance (Gs)、intercellular CO2 concentration (Ci)、transpiration rate( Tr) and the relative content of chlorophyll( SPAD) of‘ yijing12’ were greater than those of‘ xindao36’ understress condition of 125 mmol∕L NaCl. The qRT-PCR analysis revealed that both OsABCI7 and OsHCF 222 genes were significantly up-regulated in gene expression within a short period of time. The subcellular localization analysis exhibited that the protein encoded by OsABCI 7 gene was localized in chloroplasts, and the protein encoded by OsHCF 222 gene was localized in cytoplasm. Taken together, the defense ability of the photosynthetic system with the‘ yijing12’ was superior to that of‘ xindao36’, which will further provide a theoretical basis for the screening and identification of salt-tolerant rice resources.
