摘  要:目的:探讨紫龙金片联合安罗替尼对肺癌细胞新生血管生成和细胞生长的作用及其可能的作用机制。方法:将A549细胞分为对照组、紫龙金片组、安罗替尼组和联合组,分别采用甲基噻唑基四唑(MTT)法、5-乙炔基-2′-脱氧尿嘧啶核苷(Edu)法、克隆形成实验、流式细胞术、管形成实验、蛋白质免疫印迹法检测细胞存活率、增殖、凋亡、血管生成能力及相关蛋白表达情况。结果:与对照组比较,紫龙金组、安罗替尼组和联合组细胞存活率、Edu阳性细胞率、克隆形成率、管形成数量及磷酸化血管内皮生长因子受体2(p-VEGFR2)、磷酸化细胞外信号调节激酶(p-Erk)、磷酸化蛋白激酶B(p-AKT)、B细胞淋巴瘤-2蛋白(Bcl-2)均显著下降(P<0.05);同时,凋亡率及裂解型胱天蛋白酶-3(Cl-caspase-3)、Bcl-2相关X蛋白(Bax)水平均显著上升(P<0.05);分别与紫龙金组、安罗替尼组比较,联合组细胞存活率、Edu阳性细胞率、克隆形成率、管形成数量及p-VEGFR2、p-Erk、p-AKT、Bcl-2蛋白均显著增加(P<0.05),凋亡率及Cl-caspase-3、Bax蛋白水平均显著下降(P<0.05)。结论:紫龙金片可增强安罗替尼对新生血管生长和细胞增殖的抑制,并诱导癌细胞凋亡,这可能与抑制VEGFR-2磷酸化水平及其下游信号通路有关。Objective:To investigate the effect and the underlying mechanism of Zilongjin Tablets combined with Anlotinib on angiogenesis and cell growth in lung cancer cells.Methods:A549 cells were divided into control group,Zilongjin Tablets group,Anlotinib group,and Combined group.Cell survival rate,proliferation,apoptosis,angiogenesis ability,and related protein expression were detected using MTT assay,5-ethynyl-2′-deoxyuridine(EdU)assay,colony formation assay,flow cytometry,tube formation assay,and Western blot.Results:Compared with the control group,the Zilongjin Tablets group,Anlotinib group,and combined group showed significantly lower cell survival rate,EdU-positive cell rate,colony formation rate,tube formation number,and levels of phosphorylated vascular endothelial growth factor receptor 2(p-VEGFR2),phosphorylated extracellular signal-regulated kinase(p-Erk),phosphorylated protein kinase B(p-AKT),and B-cell lymphoma-2(Bcl-2)(P<0.05).At the same time,the apoptosis rate and levels of cleaved caspase-3(Cl-caspase-3)and Bcl-2-associated X protein(Bax)were significantly higher(P<0.05).Compared with the Zilongjin Tablets group and Anlotinib group,the combined group showed significantly higher cell survival rate,EdU-positive cell rate,colony formation rate,tube formation number,and levels of p-VEGFR2,p-Erk,p-AKT,and Bcl-2 protein(P<0.05),while the apoptosis rate and Cl-caspase-3 and Bax protein levels were significantly lower(P<0.05).Conclusion:Zilongjin Tablets enhance the inhibitory effect of Anlotinib on angiogenesis and cell proliferation,and induce cancer cell apoptosis.This may be related to the inhibition of VEGFR-2 phosphorylation levels and its downstream signaling pathways.
