摘　　要：目的:评估吴氏侗药烧伤油治疗皮肤烧伤创面的药效和抗炎、镇痛以及抑菌作用,为临床应用提供科学依据。方法:48只斯泼累格·多雷(SD)大鼠随机分为空白对照组、模型组、阳性对照组以及吴氏侗药烧伤油组。除空白对照组外,其余组构建背部Ⅱ度烧伤模型,分别给予生理盐水、湿润烧伤膏(MEBO)以及吴氏侗药烧伤油外涂。干预21 d后检测各组烧伤创面愈合率、病理形态、血清白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平及肝肾功能。60只KM小鼠和30只SD大鼠分层随机为空白对照组、阳性对照组以及吴氏侗药烧伤油组,雌雄各半,通过耳、足趾肿胀和醋酸扭体实验评价抗炎镇痛作用;抑菌实验评价抑菌作用。结果:与模型组比较,吴氏侗药烧伤油治疗后能促进烧伤创面愈合(P<0.01,P<0.001),改善创面组织病理形态,降低血清IL-6和TNF-α水平(P<0.05,P<0.01,P<0.001),并对肝肾功能无影响;与空白对照组比较,吴氏侗药烧伤油能显著抑制耳、足趾肿胀(P<0.05,P<0.001),降低扭体次数(P<0.001),提高镇痛率(P<0.01)和延长潜伏时间(P<0.05);吴氏侗药烧伤油对铜绿假单胞菌(PAE)、表皮葡萄球菌(SEP)具有抑制作用,对金黄色葡萄球菌(STA)无效。结论:吴氏侗药烧伤油具有促进烧伤创面愈合和抗炎、镇痛以及抑菌作用。Objective:To evaluate the therapeutic effects,anti-inflammatory,analgesic,and antibacterial properties of WU′s Dong medicine burn oil in treating skin burn wounds,providing scientific evidence for its clinical application.Methods:Forty-eight Sprague-Dawley(SD)rats were randomly divided into blank control group,model group,positive control group,and WU′s Dong medicine burn oil group.All groups except the blank control group were subjected to a second-degree burn model on the back,and treated with saline,moist exposed burn ointment(MEBO),or WU′s Dong medicine burn oil topically.After 21 days of intervention,burn wound healing rate,pathological morphology,serum interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)levels,and liver and kidney functions were evaluated.Sixty KM mice and thirty SD rats,evenly divided by gender,were randomly assigned to blank control group,positive control group,and WU′s Dong medicine burn oil group,and were tested for anti-inflammatory and analgesic effects using ear and paw swelling and acetic acid writhing assays,as well as antibacterial activity.Results:Compared with the model group,WU′s Dong medicine burn oil significantly promoted burn wound healing(P<0.01,P<0.001),improved tissue morphology,reduced serum IL-6 and TNF-αlevels(P<0.05,P<0.01,P<0.001),and had no effect on liver and kidney functions.Compared with the blank control group,WU′s Dong medicine burn oil significantly inhibited ear and paw swelling(P<0.05,P<0.001),reduced the number of writhes(P<0.001),increased pain relief rate(P<0.01),and prolonged the latency period(P<0.05).WU′s Dong medicine burn oil inhibited Pseudomonas aeruginosa(PAE)and Staphylococcus epidermidis(SEP),but had no effect on Staphylococcus aureus(STA).Conclusion:WU′s Dong medicine burn oil promotes burn wound healing and possesses anti-inflammatory,analgesic,and antibacterial effects.
