摘 要:软蜡蚧亚科Coccinae,隶属于半翅目Hemiptera,蚧次目Coccomorpha,蜡蚧科Coccidae,大多数种类是危害严重的经济害虫.以软蜡蚧亚科昆虫10属16种为研究对象,采用遗传距离分析、系统发育分析和自动条形码间隙检测方法(ABGD)评估COⅠ基因在中国软蜡蚧亚科物种界定中的适用性和可行性.结果显示:软蜡蚧亚科昆虫的种间最小遗传距离大于种内最大遗传距离,且COⅠ基因序列存在一个明显的条形码间隙;系统发育结果显示软蜡蚧亚科昆虫所有同一物种的不同样本都以高支持度聚为一支;ABGD物种界定结果显示初始划分结果与形态学鉴定结果一致;因此COⅠ基因对软蜡蚧亚科昆虫具有较高的物种识别能力,可作为该类群鉴定的有效分子标记.研究结果为识别软蜡蚧种类提供了快速准确的鉴定方法,并为研究蚧虫分类和演化等问题提供数据支持.Coccinae belongs to Hemiptera,Coccomorpha,Coccidae,Most of them are serious economic pests.In this study,16 species of 10 genera of Coccinae were used to evaluate the applicability and feasibility of COⅠ gene in the species delimitation of Coccinae from China by genetic distance,phylogeny and automatic barcode gap discovery(ABGD)method.The results showed that the minimum interspecific genetic distance of Coccinae insects was greater than the maximum intraspecific genetic distance,and there was an obvious barcode gap in the COⅠ gene sequences.Phylogenetic results showed that different samples of the same species of Coccinae insects were clustered into one branch with high support.ABGD showed that the initial partition results were consistent with the morphological identification results.Therefore,the COⅠ gene had a high species recognition ability to Coccinae insects and could be used as an effective molecular marker for the identification of this taxon.These findings offer a rapid and accurate identification method for the identification of soft scale species,and provide data support for taxonomy and evolution of scale insect.
