摘  要:为优化大孔树脂分离连翘酯苷A和连翘苷的纯化工艺,建立了含量测定方法。以静态吸附、解析性为考查指标,对5种大孔树脂进行筛选,以单因素试验考查上样浓度和洗脱流速、洗脱剂浓度、体积分数及洗脱剂用量对吸附解析性能的影响。得到连翘酯苷A和连翘苷的最佳纯化工艺为D101型大孔吸附树脂作吸附剂,药材-树脂配比1.5∶1(g∶g),上样流速2 BV/h,之后以10 BV的30%乙醇溶液和6 BV的50%乙醇溶液连续洗脱,洗脱流速为3 BV/h。To establish a HPLC method for the content determination of forsythoside A and phillyrin of Forsythiae Fructus,and to optimize the purification technology by macroprous resins.The procedure of HPLC—DAD was performed on the mobile phase.5 kinds of macroporous resins were screened out by static adsorption and desorption tests;And the flow rate,eluent concentration,volum and flow rate on the adsorption and desorption efficiences was investigated by single by single factor test.Optimum chromatogrphic on this adsorbent were obtained.D101 resin was selected as the best one,sample to resin ratio 1.5∶1(g∶g),sample loading flow flow rate 2 BV/h,sequential elution 10 BV of 30%ethanol and 6 BV of 50%ethol at a flow rate of 3 BV/h.
