摘  要:为提高茄病镰刀菌产环孢素A(1)的产量,该研究通过单因素试验和Box-Behnken设计结合响应面法优化了1发酵培养基中的关键成分,包括碳源、有机氮源和氨基酸的种类及含量。优化配方为:葡萄糖31.3 g/L,麦芽糊精62.7 g/L,酪蛋白17.3 g/L,酵母浸膏LM8018.7 g/L,L-缬氨酸7.15 g/L。在此条件下,1摇瓶产量提高至4283 mg/L。继而考察不同补料工艺对1产量的影响。最终,采用流加补料的方式使25 L发酵罐中1的产量达到5247 mg/L,相较于原始配方提高了72.49%,为1的工业化生产提供了一定参考。To improve the yield of cyclosporin A(1)produced by Fusarium solani,this study optimized the types and amounts of the key components in the fermentation medium for 1 production,including the carbon source,organic nitrogen source,and amino acid by single factor experiment and Box-Behnken design coupled with response surface methodology.The optimized formulation was as follows:glucose 31.3 g/L,maltodextrin 62.7 g/L,casein 17.3 g/L,yeast extract LM8018.7 g/L,L-valine 7.15 g/L.Under these conditions,the yield of 1 by shake flask culture was increased to4283 mg/L.Then,the effects of different supplementation processes on the yield of 1 were investigated.Finally,the yield of 1 reached to 5247 mg/L in a 25 L fermentor using fluid-feeding,which was 72.49%higher than that of the original formulation.This study provided some references for the industrial production of 1.
