摘 要:为优化紫草色素的提取工艺,以紫草为原料,采用乙醇水溶液作为提取液,以紫草素提取率为指标,通过探究提取温度、提取时间、提取液体积分数和料液比等因素对紫草素提取率的影响,采用响应面法优化紫草色素提取工艺,并进行了抗菌和抗氧化测试。结果表明,单因素中最优提取条件为:提取温度60℃,提取时间40 min,提取液体积分数90%,料液比1∶10,最大提取率2.235%。优化所得最优提取参数为:提取温度68℃,提取时间44 min,提取液体积分数90%,可得到紫草素最大提取率为2.496%。当紫草色素样品质量浓度为0.05、0.1、0.2、0.4、0.8 mg/mL时,紫草色素的抗氧化能力随着样品质量浓度的增加而提高。抗氧化能力可达83.6%,说明紫草色素具有一定清除DPPH自由基的能力。紫草色素在大肠埃希菌和金黄色葡萄球菌中均有明显的抑菌圈,说明紫草色素有一定的抗菌性。In order to optimize the extraction process of comfrey pigment,comfrey is used as raw material,ethanol aqueous solution is used as the extraction solution,the extraction rate of comfrey is taken as the index,and the effects of single factor experiments such as extraction temperature,extraction time,extraction liquid integral number and solid-liquid ratio on the extraction rate of comfrey are explored.Response surface method is used to optimize the extraction process of comfrey pigment,and antibacterial and antioxidant tests are conducted.The results show thatthe optimal extraction conditions among single factors are extraction at 60℃for 40 min with extraction solution of 90%v/v and solid-liquid ratio of 1∶10,and the maximum extraction rate is 2.235%.The optimal extraction parameters obtained through optimization are extraction at 68℃for 44 min with extraction solution of 90%v/v,and the maximum extraction rate of comfrey pigment is 2.496%.When the concentration of comfrey pigment is 0.05,0.1,0.2,0.4 and 0.8 mg/mL,the antioxidant capacity of comfrey pigment increases with the increase of the mass concentration of the sample,and the antioxidant capacity can reach 83.6%,indicating that comfrey pigment has a certain ability to scavenge DPPH free radicals.Comfrey pigment has obvious inhibitory zones in Escherichia coli and Staphylococcus aureus,indicating that comfrey pigment has certain antibacterial properties.
