基于离子交换法的戒烟贴中尼古丁的高效提取

摘　　要：尼古丁是烟草中的主要成瘾性成分,急性摄入会威胁使用者的生命安全。戒烟贴是一种基于尼古丁替代疗法的辅助戒烟产品,但目前尚未被列入烟草制品监管范围,部分产品的尼古丁含量标注不准确甚至无标识,对使用者产生严重安全隐患。戒烟贴的多层结构和复杂基质成分导致常规固液萃取法难以充分提取其中的尼古丁,这对戒烟贴中尼古丁的精确定量提出了挑战。因此,研究从戒烟贴中高效提取尼古丁的方法具有重要实践意义。根据尼古丁的结构特点,选用732阳离子交换树脂对戒烟贴中的尼古丁进行提取,并通过高效液相色谱法对提取效率进行了表征。结果表明,732阳离子树脂对游离尼古丁的吸附率为96.6%~99.3%;以甲醇与1M氢氧化钠溶液的混合液(V∶V=3∶7)对离子树脂进行洗脱,尼古丁洗脱率为87.1%~99.2%。该方法对4种不同底布材质的戒烟贴中的尼古丁综合提取率为86.03%~98.32%,实现了针对戒烟贴中的尼古丁的高效提取,为戒烟贴中的尼古丁的精确定量提供了技术支持。Nicotine is the main addictive component in tobacco,while acute intake of large quantities is life-threatening to users.Smoking cessation patch is a kind of ancillary product based on nicotine replacement therapy,but it has not been included in the scope of tobacco products.The nicotine contents of some products are not accurate or even unlabeled,which leads to serious safety risks.Because of the multilayer structure and complex chemical compositions of smoking cessation patches,it is difficult to fully extract nicotine by conventional methods,bringing a challenge to accurately quantify nicotine.Therefore,it is of great significance to develop an efficient method to extract nicotine from smoking cessation patches.According to the structural characteristics of nicotine,an extraction method was developed by 732 cationic resin and the extraction efficiency was characterized by HPLC.Results showed that the adsorption rate of 732 cationic resin for nicotine was 96.6%~99.3%,and the elution rate was 87.1%~99.2%with the mixed solution of methanol and 1M NaOH aqueous solution(V∶V=3∶7).In conclusion,an efficient extraction method was developed with the comprehensive extraction rates between 86.03%~98.32% in 4 kinds of smoking cessation patches with different substrate materials,which provided technical support for the accurate quantitation of nicotine in smoking cessation patches.
