摘 要:目的考察强壮灵合剂的镇痛、耐缺氧及抗疲劳作用。方法通过醋酸致小鼠扭体实验、热水缩尾实验、热板实验测定小鼠对不良刺激的痛阈值,醋酸致炎后测定小鼠体内环氧酶-2(cyclooxygenase-2,COX-2)的含量;通过常压耐缺氧实验、亚硝酸钠中毒实验和急性脑缺血缺氧实验,考察强壮灵合剂的耐缺氧作用;通过小鼠负重游泳实验及测定疲劳小鼠血清尿素氮、血乳酸、肝糖原、肌糖原,考察强壮灵合剂的抗疲劳作用。结果与正常对照组比较,小鼠给予强壮灵合剂后,醋酸所致小鼠扭体潜伏期显著延长,扭体次数减少,缩尾潜伏期明显延长,热板实验中给药组小鼠舔后足时间延长;醋酸致痛3 d后,小鼠血清中COX-2含量明显升高,给予强壮灵合剂后可抑制COX-2含量的升高;小鼠给予强壮灵合剂后,常压缺氧条件下的存活时间延长,亚硝酸盐中毒和急性脑缺血、缺氧小鼠的存活时间延长;与正常对照组比较,给药组小鼠力竭游泳时间延长,疲劳小鼠血清尿素氮含量降低,血中乳酸堆积下降,肝糖原、肌糖原含量增加。结论强壮灵合剂具有与阿司匹林相似但较为温和的镇痛作用,亦具有明显的耐缺氧及抗疲劳作用。Objective To investigate the analgesic,anti-hypoxia and anti-fatigue effects of Qiangzhuangling Mixture(强壮灵合剂)on mice.Methods The pain threshold of mice to adverse stimulation was determined by acetic acid induced writhing test,hot water tail shrinking test and hot plate test,and the content of cyclooxygenase-2 in mice was determined after acetic acid induced inflammation.The anti-hypoxia effects of Qiangzhuangling Mixture were investigated by normal pressure hypoxia tolerance test,sodium nitrite poisoning test and acute cerebral ischemia hypoxia test;the anti-fatigue effects of Qiangzhuangling Mixture were investigated by weight-bearing swimming test and determination of serum urea nitrogen,blood lactic acid,liver glycogen and muscle glycogen in fatigue mice.Results Compared with the normal control group,the latency of acetic acid-induced writhing was significantly prolonged,the number of writhing was reduced,the latency of tail retraction was significantly prolonged,and the time of licking hind feet was prolonged in the hot plate experiment.After 3 days of acetic acid-induced pain,the content of COX-2 in serum of mice increased significantly,which was inhibited by Qiangzhuangling Mixture.After administration of Qiangzhuangling Mixture,the survival time of mice under normal pressure and hypoxia was prolonged,and the survival time of mice with nitrite poisoning,acute cerebral ischemia and hypoxia was prolonged.Compared with the normal control group,the exhaustive swimming time of mice in the treatment group was prolonged,the content of serum urea nitrogen was decreased,the accumulation of lactic acid in blood was decreased,and the content of liver glycogen and muscle glycogen were increased.Conclusion Qiangzhuangling Mixture has a similar but mild analgesic effect to aspirin,as well as significant anti hypoxia and anti-fatigue effects.
