摘 要:目的基于数据挖掘和网络药理学分析中药处方治疗慢性咳嗽的用药规律及核心处方药物的作用机制。方法通过筛选北京中医药大学第三附属医院2020年9月—2024年1月专家门诊诊断为慢性咳嗽的患者处方信息,建立数据库,使用Excel 2019、IBM SPSS Statistics 22.0和SPSS Molder 18.0软件对数据进行分析,采用频率统计、关联规则和聚类分析等数据挖掘技术,筛选出关键的中药组合。基于中药系统药理学数据库占分析平台(TCMSP)、OMIM、GeneCards等数据库和Cytoscape软件获得关键作用靶点,进行基因本体(genes ontology,GO)功能富集分析和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)通路富集分析。结果共纳入232首处方,包含97味药物,高频中药有干姜、五味子、枇杷叶、清半夏、苦杏仁等,聚类分析可归为4类。处方包含活性成分312个,筛选药物与疾病交集靶点235个,AKT1、IL-6、VEGFA、EGFR、IL-1、MMP-9等是药物作用的关键潜在靶点。涉及生物过程937个、分子功能196个、细胞组成111个和KEGG通路177个。结论该研究初步分析中药治疗慢性咳嗽的组方用药规律及核心处方有效成分槲皮素、山柰酚、β-谷甾醇、豆甾醇、异鼠李素等可作用关键潜在靶点,通过IL-17、TNF、HIF-1、PI3K-Akt、EGFR、病毒感染、Toll样受体信号通路等发挥抗炎、抑菌、调控免疫、降低血管损害等作用,体现了中药复方治疗慢性咳嗽多成分、多位点、多通路的联合作用特点,为后续研究提供理论基础。Objective To analyze the medication pattern of traditional Chinese medicine prescription for chronic cough and the mechanism of action of core prescription drugs based on data mining and network pharmacology.Methods By screening the prescription information of patients diagnosed with chronic cough in the specialist outpatient clinic of the third affiliated hospital of Beijing university of Chinese medicine from September 2020 to January 2024,a database was established,and the data were analyzed using the software of Excel 2019,IBM SPSS Statistics 22.0,and SPSS Molder 18.0,and the data were analyzed using the data mining techniques such as frequency statistics,association rules and clustering analysis and other data mining techniques to screen out the key TCM combinations.Based on the TCMSP,OMIM,GeneCards and other databases of TCM system pharmacology analysis platform and Cytoscape software,key action targets were obtained,and GO function enrichment analysis and KEGG pathway enrichment analysis were performed.Results A total of 232 prescriptions containing 97 drugs were included,and the high-frequency traditional Chinese medicines included Ganjiang(Zingiberis Rhizoma),Wuweizi(Schisandrae Chinensis Fructus),Pipaye(Eriobotryae Folium),Qingbanxia(Rhizoma Pinelliae Preparata),and Kuxingren(Armeniacae Semen Amarum),which could be categorized into 4 classes by cluster analysis.The prescriptions contained 312 active ingredients,and 235 targets for drug-disease intersection were screened,with AKT1,IL-6,VEGFA,EGFR,IL-1,MMP-9,etc.being the key potential sites for drug action.937 biological processes,196 molecular functions,111 cellular compositions and 177 KEGG pathways were involved.Conclusion In this study,we preliminarily analyzed the prescription and dosing pattern of TCM for the treatment of chronic cough and the core prescription active ingredients quercetin,kaempferol,β-sitosterol,stigmasterol,isorhamnetin,etc.,which can act on the key potential targets and play the roles of anti-inflammatory,anti-bacterial,immune-
