摘 要:目的探讨携IL-6和IL-8抗体靶向超声微泡评价兔心肌缺血/再灌注损伤(MIRI)的效果。方法144只兔随机分为3组:缺血/再灌注组(I/R组)96只、假手术组(S组)24只、正常组(N组)24只。以上各组又随机平均分为:MBc组、MB_(IL-6)组、MB_(IL-8)组及MBd组。构建携IL-6和IL-8单抗及双抗靶向微泡。心肌声学造影分析前壁的视频强度(video intensity,VI),将VI与血清中IL-6和IL-8的水平做相关性分析。结果成功构建携单抗及双抗靶向微泡。靶向微泡:I/R组兔前壁VI明显高于S组和N组,且随着再灌注(Rep)时间的延长,VI逐渐升高,MB_(IL-6)和MBd于Rep 180 min达高峰,MB_(IL-8)于Rep 120 min和Rep 180min达高峰。相同Rep时间,4种微泡VI:MBd>MB_(IL-8)>MB_(IL-6)>MBc。靶向微泡I/R组前壁VI与IL-6与IL-8水平均呈正相关(均r>0.7,P<0.05)。结论携双抗靶向微泡比携单抗靶向微泡对MIRI的显像灵敏度更高、靶向性更强,可实现无创和定量评价MIRI的炎症反应程度。Objective To explore the effect of targeting ultrasound microbubbles with IL-6 and IL-8 antibodies on evaluating myocardial ischemia/reperfusion injury(MIRI)in rabbits.Methods A total of 144 rabbits were randomly divided into three groups:96 rabbits in the ischemia/reperfusion group(I/R group),24 rabbits in the sham operation group(S group)and 24 rabbits in the normal group(N Group).The above groups were randomly sub-divided into MBc group,MB_(IL-6)group,MB_(IL-8)group,and MBd group.Myocardial contrast echocardiography was used to analyze the video intensity(VI)of the anterior wall,and the correlation between VI and the levels of IL-6 and IL-8 in serum was analyzed.Results Targeted microbubbles with monoclonal antibodies and double antibodies were successfully constructed.Targeted microbubbles:The anterior wall VI of rabbits in the I/R group was significantly higher than that in the S group and the N groups,and gradually increased with the prolongation of reperfusion(Rep)time.MB_(IL-6)and MBd reached their peak at Rep 180 min,while MB_(IL-8)reached its peak at Rep 120 min and Rep 180 min.At the same Rep time,four types of microbubbles VI:MBd>MB_(IL-8)>MB_(IL-6)>MBc.The anterior wall VI of the targeted microbubble I/R group was positively correlated with the levels of IL-6 and IL-8(all r>0.7 and P<0.05).Conclusions Targeted microbubbles with double antibodies are more sensitive and targeted to MIRI than targeted microbubbles with monoclonal antibodies,which can achieve non-invasive and quantitative evaluation of the degree of inflammatory response of MIRI.
