摘 要:通过对患水霉病泰山螭霖鱼病原菌进行分离鉴定,并研究其生物学特性,以期为泰山螭霖鱼水霉病的防治提供科学依据。运用传统方法从泰山螭霖鱼体表病灶处分离纯化出一株优势水霉菌株,命名为TS01。经形态学观察可见,其菌丝细长无横隔,游动孢子在孢子囊内多排有序排列,自顶端释放后迅速向四周游走。通过与GenBank中已知序列比较,发现TS01菌株水霉属亲缘关系最近,同源性高达99%。结合形态学特征及系统发育鉴定结果,可判定TS01菌株为澳大利亚水霉(Saprolegnia australis)。利用单因素实验法,研究不同生长条件对菌株TS01生长速率的影响,发现菌株TS01在10℃~30℃、pH5~11条件下均可以生长,最适温度为25℃,最适pH为8。此外,研究还表明NaCl浓度的升高对菌株TS01的生长具有明显的抑制作用,在2%NaCl浓度下,菌株TS01几乎不生长。本研究完成了对泰山螭霖鱼水霉病病原菌TS01的鉴定,详细描述了其生物学特性,进一步丰富了澳大利亚水霉的流行范围,为螭霖鱼水霉病的防治提供了理论数据支持。This research was designed to isolate and identify the pathogen of Varicorhinus macrolepis suffering from saprolegniasis,and investigate its biological characteristics to provide a scientific basis for the prevention and control of saprolegniasis in Varicorhinus macrolepis.A dominant strain TS01,was isolated and purified from the lesion sites on the body surface of Varicorhinus macrolepis using the traditional method.Morphological observations revealed that its hyphae were slender without cross-septa,and the zoospores were orderly arranged in multiple rows within the sporangia,rapidly moving around after being released from the apex.By comparing the ITS rDNA sequence of the TS01 strain in GenBank,it was found that the ITS rDNA sequences of strain TS01 was naturally clustered with ITS rDNA sequences of Saprolegnia australis,with a homology of up to 99%.Combined morphological characterization with phylogenetic analysis based on ITS rDNA sequence,strain TS01 was identified as Saprolegnia australis.Using the single-factor experimental method,the effects of different growth conditions on the growth rate of strain TS01 were studied.It was found that strain TS01 could grow at 10℃~30℃ and pH5~11,its optimum growing temperature and pH were 25℃ and 8.In addition,the study also indicated that the increase in NaCl concentration had a significant inhibitory effect on the growth of strain TS01.Under the condition of 2%NaCl concentration,strain TS01 hardly grew at all.This study has completed the identification of the pathogen TS01 of Saprolegniasis in Varicorhinus macrolepis,described its biological characteristics in detail,further enriched the epidemic range of Saprolegnia australis,and provided theoretical data support for the prevention and treatment of Saprolegniasis in Varicorhinus macrolepis.
