摘 要:本研究利用Red同源重组系统和互补质粒构建禽致病性大肠杆菌(avian pathogenic Escherichia coli,APEC)菌株APCE50的tolQ基因缺失株ΔtolQ和互补株CΔtolQ,以探究tolQ基因对APEC生物学特性以及对细菌致病性的影响。对APCE50、ΔtolQ和CΔtolQ的生长曲线、运动能力、形态特征、生物被膜形成能力、酸碱耐受性、抗血清杀菌、细胞黏附侵袭能力及毒力进行了比较分析。结果显示,ΔtolQ与APCE50、CΔtolQ的生长速度无明显差异,但ΔtolQ的运动能力下降并影响细菌形态。缺失tolQ基因导致APEC生物被膜形成能力、酸碱耐受性及抗血清杀菌能力显著降低。细胞及动物感染试验结果显示,tolQ基因缺失显著降低了APEC的黏附侵袭能力和毒力。综上表明,tolQ基因在APEC运动能力、生物被膜形成、耐受性及致病性方面发挥作用,这为阐明APEC的环境适应性、生存能力及致病机制提供了参考。This study focused on examining the role of the tol Q gene in the biological characteristics and pathogenicity of avian pathogenic Escherichia coli(APEC).Using the Red homologous recombination system and complementation plasmid,tol Q gene mutant and complemented strains of APEC were constructed.Then,we compared the the growth patterns,motility,morphology,biofilm formation capacity,survival in acidic and alkaline environments,cell adhesion and invasion capabilities,and pathogenicity of APCE50,Δtol Q,and CΔtol Q strains.The results indicated that the mutant strain exhibited decreased motility and altered bacterial morphology.The absence of the tol Q gene led to significant decreased biofilm formation capacity,reduced survival rate in acidic and alkaline conditions,and diminished serum bactericidal resistance.Cell and animal infection assays revealed that the tol Q mutant strain exhibited significantly diminished capacity to adhere to and invade cells,as well as attenuated virulence in animal.In summary,the tol Q gene plays a significant role in APEC motility,biofilm formation,tolerance,and pathogenicity.These findings provide insights into the environmental adaptation,survival,and pathogenic mechanisms of APEC.The study contributes to our understanding of the environmental adaptability,adhesion and invasion abilities,and virulence mechanisms of APEC.
