摘 要:目的观察丹参酚酸B对创伤性脑损伤(TBI)大鼠脑组织中脑水肿及脑血流的影响。方法48只健康雄性SPF级SD大鼠随机分为假手术组,模型组,丁基对苯二酚(TBHQ,25 mg·kg^(-1))组,丹参酚酸B低、中、高剂量(7.5、15、22.5 mg·kg^(-1))组。采用脑皮质电子损伤撞击仪建立TBI模型,假手术组大鼠仅开骨窗,不做撞击。各组于制模24 h内分别灌胃给予相应剂量药物,假手术组和模型组灌胃给予生理盐水。采用干湿-重法测定脑组织含水量百分比,采用激光散斑血流成像检测脑血流量。结果与假手术组相比,模型组损伤区和对侧皮质脑组织含水量均增加,脑血流量显著减少(P<0.05)。与模型组相比,丹参酚酸B各剂量组和TBHQ组损伤区和对侧皮质脑组织含水量均减少,脑血流量显著增加(P<0.05)。丹参酚酸B高剂量组脑组织含水量和脑血流量与TBHQ组相比均无显著差异(P>0.05)。结论丹参酚酸B通过改善TBI后脑水肿、增加脑血流量灌注,发挥神经保护作用。AIM To observe the effects of salvianolic acid B on cerebral edema and cerebral blood flow in rats with traumatic brain injury(TBI).METHODS Fourty-eight healthy male SPF-grade SD rats were randomly divided into 6 groups:sham group,model group,tert-butylhydroquinone(TBHQ,25 mg·kg^(-1))group,salvianolic acid B low,medium and high dose(7.5,15,22.5 mg·kg^(-1))groups.TBI model was established by using a cerebral cortex electronic injury impactor after bone window was opened.The rats in the sham group were only opened to the bone window without impact.Each group was given the corresponding dose of TBHQ or salvianolic acid B by gavage within 24 h after TBI,and saline was given by gavage in the sham group and the model group.The water content of brain tissue was determined by wet-dry-weight method.Cerebral blood flow was detected by laser diffusion flow imaging.RESULTS Compared with the sham group,the water content of the injured area and contralateral cortical brain tissue increased and cerebral blood flow significantly decreased in the model group(P<0.05).Compared with the model group,the water content of the injured area and contralateral cortical brain tissue was decreased and cerebral blood flow was significantly increased in all doses of salvianolic acid B groups and TBHQ group(P<0.05).There were no significant differences between the salvianolic acid B high dose group and the TBHQ group(P>0.05).CONCLUSIOVN Salvianolic acid B exerts neuroprotective effects after TBI improving cerebral edema and increasing cerebral blood flow perfusion.
