柑橘黄龙病侵染对“赣南早”脐橙光合色素、酶及矿质元素的影响

摘  要:为探明“赣南早”脐橙受柑橘黄龙病侵染后体内光合色素、酶活性及矿质元素含量的变化,进而为了解黄龙病侵染对寄主造成的影响和致病机理提供参考,对1年生树通过嫁接染病接穗获得染病植株,以嫁接健康接穗为对照,12个月后开展PCR检测验证,电镜扫描观察叶片组织结构,测定叶片光合色素、丙二醛(MDA)含量、过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性及矿质元素含量。结果表明,受黄龙病侵染的“赣南早”脐橙植株叶片气孔凸起,周围组织皱缩,叶片中脉石细胞和薄壁细胞排列松散,叶片叶绿素a、叶绿素b及类胡萝卜素含量分别是对照(健康)植株叶片的66.31%、76.42%和78.33%,叶片MDA含量增加了51.77%,POD、CAT和SOD活性分别提升了21.96%、15.43%和13.85%,叶片N、Mg和Ca含量分别减少28.11%、9.77%、13.95%,B、Cu、Mn、Fe、Zn和Mo含量减少值在6.23%~27.83%之间,K和P含量无显著变化。柑橘黄龙病侵染“赣南早”脐橙会导致寄主光合作用、呼吸作用及蒸腾作用、矿质元素吸收利用等生理功能减弱,进而影响植株正常生长发育。In order to investigate the changes of photosynthetic pigments,enzyme activities and mineral element contents in Gannanzao navel oranges infected with citrus Huanglongbing(HLB),and to provide reference for understanding the effects of HLB infection on the hosts and the pathogenic mechanism,the infected plants were obtained by grafting infected scion on one-year-old trees,and the grafted with healthy scion was used as the control.After 12 months,the PCR assay was performed for validation.The tissue structure of leaves was observed by electron microscope scanning,the photosynthetic pigments,malondialdehyde(MDA)content,the activities of peroxidase(POD),catalase(CAT),superoxide dismutase(SOD),and the mineral element contents were determined.The results showed that the leaves of infected Gannanzao navel orange plants with HLB had raised stomata,with wrinkled surrounding tissues,loosely arranged veinstone cells and thin-walled cells,and the content of chlorophyll a,chlorophyll b,and carotenoids in leaves was 66.31%,76.42%,and 78.33%higher than that of the control(healthy)plants,respectively.Leaf MDA content increased by 51.77%,and the activity of POD,CAT and SOD was increased by 21.96%,15.43%and 13.85%,respectively,while the content of N,Mg and Ca in leaves was reduced by 28.11%,9.77%and 13.95%,respectively,and the amount of B,Cu,Mn,Fe,Zn and Mo decreased by 6.23%to 27.83%,while K and P contents did not change significantly.Infection of Gannanzao navel oranges by HLB can lead to the weakening of physiological functions of the host such as photosynthesis,respiration and transpiration,as well as the absorption and utilization of mineral elements,which in turn will affect the normal growth and development of the plant.
