摘 要:开展茄梨HY5基因克隆及生物信息学分析,可为茄梨HY5基因功能的研究奠定基础。以茄梨为试材,提取mRNA,反转录为cDNA,设计特异性引物(F:ATGCAAGAGCAGGCGACG,R:TCAATCCGCATTTGCACT)扩增HY5基因全长序列,并进行生物信息学分析。结果显示,茄梨HY5基因(PcHY5)全长为495 bp,编码164个氨基酸;PcHY5蛋白质分子量为17.90 KD,为亲水蛋白质,具有bZIP结构域;PcHY5与云南红梨(红梨1号)、白梨等蔷薇科植物的HY5基因具有高度的同源性,PcHY5编码的蛋白质与中国白梨等的HY5基因编码的蛋白质相似性高达99.8%。表明,PcHY5与梨属其他植物的HY5基因具有相同功能。Cloning and bioinformatics analysis of HY5 gene of Clapp’s Favorite were carried out in order to lay a foundation for the study of gene function.The mRNA of Clapp’s Favorite pear was extracted,reverse transcribed into cDNA,and the full-length sequence was amplified by specific primers(F:ATGCAAGAGCAGGCGACG,R:TCAATCCGCATTTGCACT),and the bioinformatics of Clapp’s Favorite were analyzed.The results showed that the total length of HY5 gene was 495 bp,encoding 164 amino acids.The molecular weight of PcHY5 protein was 17.90 KD,and it was a hydrophilic protein with bZIP domain.The HY5 gene had a high homology with that of Yunnan red pear,white pear and other Rosaceae plants,and the similarity of HY5 encoded protein with that of Chinese white pear was as high as 99.8%,indicating that PcHY5 had the same function with the HY5 genes of other plants of Pyrus.
