摘 要:本研究通过比较带内生真菌(E+)、不带内生真菌(E-)和带菌率为50%(M)的苇状羊茅(Festuca arundinacea)在不同温度条件下的生理和表型变化,探究内生真菌在不同温度条件下对苇状羊茅内部生理机制、生长和抗寒性的影响。结果表明:1)25℃(常温)条件下苇状羊茅在E+处理与E-处理下的抗坏血酸过氧化物酶(Ascorbate peroxidase,APX)、过氧化物酶(Peroxidase,POD)活性和脯氨酸含量无显著差异。2)随着温度降低,E+处理的APX、POD、超氧化物歧化酶(Superoxide dismutase,SOD)和过氧化氢酶(Catalase,CAT)活性,以及脯氨酸、可溶性糖、可溶性蛋白含量均显著高于E-处理(P<0.05);0℃时,E+处理比E-处理的上述七项生理指标分别提升了14.2%、88.6%、38.0%、63.3%、54.8%、62.8%、11.0%。3)在5℃恒温培养条件下,E+处理的株高和叶长始终显著高于E-处理(P<0.05),培养60 d后E+处理的分蘖数和鲜草产量显著高于M和E-处理(P<0.05)。研究表明低温胁迫下内生真菌可以提高苇状羊茅抗氧化酶活性和渗透调节物质含量,促进苇状羊茅生长,增强其抗寒能力。In this research,the physiological and phenotypic changes of Festuca arundinacea with endophytic fungi(E+),without endophytic fungi(E-)and with a carrier rate of 50%(M)were compared under different temperature conditions.Effects of endophytic fungi on the internal physiological mechanism,growth,and hardiness of Festuca arundinacea under different temperature conditions were explored.Results revealed that under 25℃(room temperature),there was no significant difference in ascorbate peroxidase(APX)and peroxidase(POD)activities and proline content between E+and E-treatments of Festuca arundinacea.With the decrease of temperature,the activities of APX,POD,superoxide dismutase(SOD)and catalase(CAT),as well as the contents of proline,soluble sugar and soluble protein in E+treatment were significantly higher compared to E-treatment(P<0.05).At 0℃,compared to E-treatment,the seven physiological indices of E+treatment were increased by 14.2%,88.6%,38.0%,63.3%,54.8%,62.8%,and 11.0%,respectively.When cultivated under constant temperature of 5℃,the plant height and leaf length of E+treatment were always significantly higher than those of E-treatment(P<0.05).After 60 days of cultivation,the tiller number and fresh grass yield of Festuca arundinacea in E+treatment were significantly higher compared to M and E-treatments(P<0.05).This study indicated that under low temperature stress,endophytic fungi can improve the antioxidant enzyme activity and osmotic regulatory substance content,promote the plant growth,and enhance the hardiness of Festuca arundinacea.
