摘 要:血小板衍生生长因子受体B(PDGFRB)是一种细胞表面的酪氨酸激酶受体,不仅调节细胞增殖、迁移和存活过程,还对组织修复、血管形成、生殖和氧化应激具有调控作用。为探究PDGFRB与牦牛隐睾发生及隐睾氧化应激的关系,在前期转录组测序的基础上,以健康牦牛的睾丸和隐睾组织为研究材料,通过苏木精-伊红(H&E)染色观察睾丸组织和隐睾组织的形态结构,通过生物信息学分析筛选出与生殖及氧化应激相关的候选基因PDGFRB。利用实时荧光定量PCR(qRT-PCR)、蛋白免疫印迹(Western blot)和间接免疫荧光(IF)等方法检测PDGFRB的mRNA和蛋白表达及分布情况,探究PDGFRB与隐睾发生的关系。H&E结果显示,正常睾丸组织基底膜和间质组织结构完整,排列紧密,隐睾中生精小管断裂萎缩,间质细胞紧密连接受损。qRT-PCR结果显示,与正常睾丸相比,隐睾中PDGFRB mRNA的表达水平极显著升高(P<0.01)。Western blot结果显示,PDGFRB在睾丸和隐睾组织中均有表达,且隐睾中的蛋白表达水平显著高于正常睾丸组织(P<0.05)。IF结果显示,PDGFRB在睾丸支持细胞、间质细胞及生精细胞中均有表达。本研究揭示了PDGFRB可能参与隐睾的发生与氧化应激过程,为解析隐睾的发生过程提供了理论依据。Platelet-derived growth factor receptor B(PDGFRB),a tyrosine kinase receptor located on the cell surface,is not only involved in the regulation of cell proliferation,migration and survival,but also in the regulation of tissue repair,angiogenesis,reproduction and the response to oxidative stress.In order to explore the relationship between PDGFRB and the development of cryptorchidism,as well as the oxidative stress of cryptorchidism in yaks,the normal testis and cryptorchidism tissue from yaks were used as study material,based on our previous transcriptome sequencing.Hematoxylin-eosin(H&E)staining was used to observe the morphological structure of normal testis and cryptorchidism tissue,and bioinformatics analysis was used to screen out the candidate target gene PDGFRB,which is related to yak reproduction and oxidative stress.For exploring the relationship between PDGFRB and cryptorchidism,quantitative real-time PCR(qRT-PCR),Western blot and indirect Immunofluorescence(IF)were used to detect the mRNA or protein expression and distribution of PDGFRB.The H&E results showed that the basement membrane and interstitial tissue of normal testis were structurally intact and tightly aligned,while the seminiferous tubules in cryptorchidism were disrupted and atrophied,and the compact junctions of the Leydig cells were damaged.The qRT-qPCR results showed a significant increase in the expression levels PDGFRB in cryptorchidism compared to the normal testis(P<0.01).Western blot results showed that PDGFRB was expressed in both the testis and cryptorchidism,with higher expression in cryptorchidism(P<0.05).IF results showed that PDGFRB was expressed in Sertoli cells(SCs),Leydig cells(LCs)and spermatogonium.This study revealed that PDGFRB may be involved in the occurrence and oxidative stress of cryptorchidism,which provides a theoretical basis for exploring the process of cryptorchidism.
