青芪胶囊对溃疡性结肠炎小鼠M2型巨噬细胞的影响

摘  要:目的验证青芪胶囊可以促进M2型巨噬细胞极化,探究青芪胶囊治疗溃疡性结肠炎的可能作用机制。方法将40只SPF级雄性C57小鼠,随机分为空白组、对照组(美沙拉嗪缓释颗粒灌胃)、干预组(青芪胶囊)和模型组,每组10只。采用葡聚糖硫酸钠(DSS)诱导小鼠溃疡性结肠炎,对照组选美沙拉嗪缓释颗粒灌胃,干预组选靑芪胶囊中药粉灌胃,模型组灌胃给予同体积蒸馏水。采用结肠病理损伤评分、实时荧光定量PCR(RT-qPCR)检测小鼠结肠组织IL-10、TGF-β1、Arg-1的mRNA表达、流式细胞分析技术检测结肠中M2型巨噬细胞(F4/80、CD206)比例。结果通过观察小鼠结肠病理组织,发现对照组和干预组,能明显修复上皮细胞,减少炎症细胞浸润(P<0.05)。模型组中TGF-β1mRNA表达有显著升高(P<0.01),对照组与干预组TGF-β1 mRNA表达有明显降低(P<0.01)。模型组IL-10、Arg-1的小鼠结肠组织mRNA表达有显著降低(P<0.01),干预组和对照组IL-10、Arg-1的小鼠结肠组织mRNA表达有显著升高(P<0.01或P<0.05)。模型组小鼠结肠组织M2巨噬细胞(F4/80、CD206)比例明显下降(P<0.01)。干预组和对照组M2巨噬细胞(F4/80、CD206)比例明显上升(P<0.01)。结论青芪胶囊能有效减轻DSS诱导的小鼠溃疡性结肠炎,可能与抑制TGF-β1的表达,促进IL-10、Arg-1、CD206表达,从而缓解炎症反应,修复肠粘膜有关。Objective:To verify the ability of Qingqi Capsules to promote M2 macrophage polarization and explore their potential mechanisms in the treatment of ulcerative colitis(UC).Methods:Forty male SPF-grade C57 mice were randomly divided into four groups:blank group,control group(treated with mesalazine sustained-release granules via gavage),intervention group(treated with Qingqi Capsule powder via gavage),and model group(treated with an equal volume of distilled water via gavage),10 mice per group.Ulcerative colitis was induced by sodium dextran sulfate(DSS)in the mice.The control group was given continuous release granules,the intervention group was given powder of Qingqi Capsule,and the model group was given the same volume of distilled water.Colon pathological damage scores and real-time quantitative fluorescent PCR(RT-qPCR)were used to detect the mRNA expression of IL-10,TGF-β1 and Arg-1 in mouse colon tissues,and the flow cytometry was used to detect the proportion of M2 macrophages(F4/80,CD206)in the colon tissues.Results:Histopathological analysis revealed significant epithelial repair and reduced inflammatory cell infiltration in the control and intervention groups compared to the model group(P<0.05).TGF-β1 mRNA expression was significantly elevated in the model group(P<0.01)while it was markedly reduced in the control and intervention groups(P<0.01).IL-10 and Arg-1 mRNA expression in the colon tissues was significantly reduced in the model group(P<0.01)but significantly increased in the control and intervention groups(P<0.01 or P<0.05).The proportion of M2 macrophages(F4/80,CD206)was significantly lower in the model group(P<0.01),whereas it was significantly higher in the control and intervention groups(P<0.01).Conclusion:Qingqi Capsules effectively mitigate DSS-induced UC in mice,potentially through inhibiting TGF-β1 expression and promoting IL-10,Arg-1,and CD206 expression,which contributes to alleviating inflammation response and repairing intestinal mucosa.
