摘 要:目的优化复方甘芪止咳颗粒组方的提取工艺。方法浓氨水引咳法建立小鼠咳嗽模型;高效液相色谱法测定甘草酸铵含量;以乙醇浓度、溶剂用量和提取时间为影响因素,以得膏率、甘草酸铵含量、引咳潜伏时间为评价指标,采用正交实验对复方甘芪止咳颗粒组方的提取工艺进行优化。结果以60%乙醇为溶剂,溶剂体积为药材质量的4倍,煎煮4 h得到的提取物综合评分最高,为复方甘芪止咳颗粒组方的优化提取工艺。结论该提取工艺合理可行,得膏率和甘草酸铵提取率较高,引咳潜伏期长。Objective To optimize the extraction process of the formulation of Compound Ganqi Anti-Cough Granules.Methods A cough model was established in mice using the method of concentrated ammonia induced cough.The content of ammonium glycyrrhizinate was determined by high performance liquid chromatography.The concentration of ethanol,amount of solvent and extraction time were taken as the influencing factors.The extract yielding rate,content of ammonium glycyrrhizinate and the latent period of inducing cough were taken as the evaluation indexes.Results The extract obtained by decocting for 4 hours with 60%ethanol as the solvent and the volume of solvent being 4 times the mass of herbs had the highest overall score,which was the optimized extraction process of Compound Ganqi Anti-Cough Granules.Conclusion The extraction process is reasonable and feasible,with high extract yielding rate,high extraction rate of ammonium glycyrrhizinate,and a long latent period of inducing cough.
