藏药裂叶独活的小鼠急性毒性实验研究

摘  要:目的:研究药食同源裂叶独活的总提取物进行小鼠急性毒性实验,以初步评价其安全性。方法:通过预实验的给药组数与剂量,对裂叶独活总提取物进行LD 50和MTD的测试;实验组分别以80.80 g/kg、68.68 g/kg、58.38 g/kg、49.62 g/kg、42.18 g/kg的剂量进行灌胃给药,空白组以双蒸水灌胃。结果:得出裂叶独活的总提取物在不引起死亡的最大耐受量(MTD)为42.18 g/kg,半数致死量为LD 50=61.30 g/kg,发现95%置信区间为56.54~66.46 g/kg(通过改进寇氏法计算)。结论:此实验得出裂叶独活的总提取物属于微毒或无毒物质,在试验剂量范围内有较高的安全性,为国家药食同源安全使用及发展提供科学依据。Objective To conduct acute toxicity experiments on the total extract of medicinal and edible homologous Ligusticum multiflorum in mice,in order to preliminarily evaluate its safety.Method LD 50 and MTD tests were conducted on the total extract of Angelica lobata using the number and dosage of administration groups in the pre experiment;The experimental group was administered orally at doses of 80.80 g/kg,68.68 g/kg,58.38 g/kg,49.62 g/kg,and 42.18 g/kg,respectively,while the blank group was administered orally with double distilled water.Result The maximum tolerable dose(MTD)of the total extract of Angelica lobata without causing death was 42.18 g/kg,and the median lethal dose was LD 50=61.30 g/kg.It was found that the 95%confidence interval was 56.54 g/kg to 66.46 g/kg(calculated by improved Kou's method).Conclusion This experiment concluded that the total extract of Lonicera lobata belongs to the category of slightly toxic or non-toxic substances,and has high safety within the experimental dose range,providing scientific basis for the safe use and development of medicinal and food homology in the country.
