摘 要:目的:利用高效液相色谱法(HPLC)探究低(0~500 m)、中(500~1000 m)、高(1000~1500 m)三个海拔无患子果皮中指标性成分常春藤皂苷元的含量差异,并对其检测方法进行分析验证。方法:DiamonsilC_(18)色谱柱(4.6 mm×250 mm,5μm);以乙腈(A)-水(B)为流动相,80%(A):20%(B)比例进行等度洗脱;流速为1.0 mL/min;柱温为30℃;检测波长为210 nm;进样体积为10μL。结果:得到常春藤皂苷元线性方程Y=330236X+302268,R^(2)=0.9993,各组平均加样回收率为98.52%,RSD=2.56%。3个海拔无患子样品中常春藤皂苷元含量有所差异,最高达到58.066 mg/g,最低仅有24.493 mg/g,中海拔和高海拔的平均含量较为接近,分别为46.121 mg/g、46.380 mg/g,均比低海拔的平均含量(42.126 mg/g)高。结论:该方法准确稳定,可用于不同海拔无患子果皮中指标性成分常春藤皂苷元的含量测定,同时为三个海拔无患子果实资源的开发利用提供了理论依据。Objective:To investigate the differences in the content of the indicator component hederagenin in the pericarp of Sapindus saponaria at three altitudes,low(0-500m),medium(500-1000m)and high(1000-1500m),using High Performance Liquid Chromatography(HPLC).Methods Diamonsil C_(18) column(4.6 mm×250 mm,5μm);isocratic elution with acetonitrile(A)-water(B)as the mobile phase in the ratio of 80%(A):20%(B);flow rate of 1.0 mL/min;column temperature of 30℃;detection wavelength of 210 nm;injection volume of 10μL.Results:The linear equation of hederagenin Y=330236X+302268 was obtained with R^(2)=0.9993.The average recoveries of each group were 98.52%with RSD=2.56%.The hederagenin contents of the samples of Sapindus saponaria from three elevations varied,with the highest reaching 58.066 mg/g and the lowest 24.493 mg/g.The average contents of samples from the middle elevations and high elevations were closer to 46.121 mg/g and 46.380 mg/g,respectively,than the average contents of 42.126 mg/g of low elevation samples.Conclusion:The method is accurate and stable,and can be used for the determination of the content of the index constituent hederagenin in the pericarp of Sapindus saponaria at different altitudes,meanwhile,it provides a theoretical basis for the development and utilisation of the resources of Sapindus saponaria fruits at three altitudes.
