摘 要:建立了一个高效、准确的高效液相串联质谱测定附子中3种生物碱的方法。样品经甲醇提取,高效液相色谱串联色谱仪检测,外标法定量。结果表明,苯甲酰新乌头碱、苯甲酰乌头原碱和苯甲酰次乌头原碱的线性范围为2~1000 ng/mL,线性相关系数不小于0.9956,方法检出限分别为0.2μg/kg、0.4μg/kg和0.4μg/kg,定量限分别为0.4μg/kg、0.75μg/kg和0.75μg/kg。添加1μg、5μg和10μg的3种生物碱的混标,苯甲酰新乌头碱、苯甲酰乌头原碱和苯甲酰次乌头原碱的回收率分别为85.1%~96.0%,98.4%~100.2%和94.7%~96.1%,相对标准偏差分别为3.90%~5.98%,3.21%~6.01%和3.66%~4.32%。该方法高效、准确地测定了中草药附子中3种生物碱含量。An efficient and accurate high performance liquid chromatography tandem mass spectrometry method for the determination of three alkaloids in Radix Aconiti Lateralis Preparata was developed.The samples were extracted by methanol,detected by high performance liquid chromatography tandem mass spectrometry and quantified by external standard method.The results showed that the linear ranges of benzoyl neoaconitine,benzoylaconitine and benzoylpseudoaconitine were 2~1000 ng/mL with the linear correlation coefficients not less than 0.9956,the detection limits were 0.2μg/kg,0.4μg/kg,and 0.4μg/kg,and limit of quantitation were 0.4μg/kg,0.75μg/kg and 0.75μg/kg respectively.After adding 1μg,5μg and 10μg of the three alkaloids,the recovery rates of benzoylneaconitine,benzoylaconitine and benzoylsubaconitine were 85.1%~96.0%,98.4%~100.2%and 94.7%~96.1%,respectively,and the relative standard deviations were 3.90%~5.98%.3.21%~6.01%and 3.66%~4.32%,respectively.The method achieved an efficient and accurate determination of three alkaloids in the Chinese herbal medicinea Radix Aconiti Lateralis Preparata.
