摘  要:为研究黄芩苷(baicalin, BA)对尿酸钠晶体(monosodium urate crystal, MSU)诱导大鼠肾小管上皮细胞(NRK-52e)损伤的影响及其机制,培养NRK-52e细胞,筛选出MSU最佳造模浓度和BA给药浓度,用试剂盒测定一氧化氮(NO)、乳酸脱氢酶(LDH)、肿瘤坏死因子(TNF-α)和白介素1β(IL-1β)的含量,用荧光探针检测细胞凋亡情况以及细胞内活性氧(ROS)水平。结果显示,黄芩苷能显著降低细胞上清液中NO、LDH、TNF-α和IL-1β的含量(P<0.05或P<0.01),减缓细胞凋亡速度和减少细胞内的ROS含量。研究表明黄芩苷可通过抗炎、抗氧化以及抗凋亡来减轻尿酸钠晶体诱导的大鼠肾小管上皮细胞损伤,可为将黄芩苷开发成治疗高尿酸血症的新药提供实验依据。In order to study the effect of baicalin(BA)on the injury of rat renal tubular epithelial cells(NRK-52e)induced by monosodium urate crystal(MSU)and its mechanism,NRK-52e cells were cultured,and the optimal concentration of MSU and BA administration concentration were screened.The contents of nitric oxide(NO),lactate dehydrogenase(LDH),tumor necrosis factor(TNF-α)and interleukin-1β(IL-1β)were determined by the kit.Apoptosis and intracellular reactive oxygen species(ROS)were detected by fluorescent probe.The results showed that baicalin could significantly reduce the contents of NO,LDH,TNF-αand IL-1βin cell supernatant(P<0.05 or P<0.01),slow down apoptosis and reduce intracellular ROS content.The results show that baicalin can reduce the damage of rat renal tubule epithelial cells induced by sodium urate crystals through anti-inflammatory,antioxidant and anti-apoptosis,which can provide experimental basis for the development of baicalin as a new drug for the treatment of hyperuricemia.
