摘  要:以川桑叶粉末为原材料,在酶提法的基础上,结合超声波辅助探究桑叶总生物碱的提取工艺。在单因素试验的基础上,通过响应面法设计优化试验,以桑叶总生物碱含量为指标,探究酶浓度、液料比、提取温度、超声时间、pH对桑叶总生物碱含量的影响以及732型阳离子交换树脂对提取物的纯化效果。结果显示,酶浓度对含量的影响最大,最佳的提取工艺条件是酶浓度为827 U/100 mL、液料比为31、pH值为5.0、超声时间为30 min、提取温度为50℃,此条件下进行提取,桑叶总生物碱的得率达到4.21±0.07 mg/g。经过阳离子交换树脂静态纯化后,桑叶生物碱DNJ的纯度由0.37%提高至2.95%。以上结果说明超声酶解法能在一定程度上提高桑叶总生物碱的提取含量,经优化后工艺条件准确可靠,且采用732树脂纯化DNJ具有可行性和稳定性。研究可为桑叶总生物碱的提取和纯化提供一定理论依据。With Sichuan mulberry leaves as raw materials,the research explored the extraction process of total alkaloids from morus alba based on enzymatic extraction combined with ultrasound assistance.First conduct a single factor experiment,then the response surface method was used to design the optimization test,and the total alkaloid was used as the index to explore the effects of enzyme concentration,liquid-solid ratio,extraction temperature,ultrasonic time and pH on the total alkaloid content of mulberry leaves and the purification effect of 732 cation exchange resin on the extract.The results showed that the enzyme concentration had the greatest influence on the content.The optimum extraction conditions were as follows:the enzyme concentration was 827 U/100 mL,the ratio of liquid to material was 31,the extraction temperature was 50℃,the ultrasonic time was 30 min,and pH value was 5.0.In this circumstance,the total alkaloids yield of mulberry leaves reached 4.21±0.07 mg/g.After the static purification by cation exchange resin,the purity of DNJ increased from 0.37%to 2.95%.The above results indicated that ultrasonic enzymatic hydrolysis can improve the extraction content of total alkaloids from mulberry leaves to a certain extent,and the optimized process conditions are accurate and reliable.The purification of DNJ using 732 resin is feasible and stable.This research provides some theoretical basis for the study of total alkaloids of mulberry leaves.
