摘 要:目的基于睾丸支持细胞线粒体自噬途径分析菟丝子中黄酮类成分对精子细胞活力所产生的影响。方法针对睾丸支持原代细胞实施传代培养,然后加入不同浓度菟丝子黄酮处理睾丸支持细胞,培养72h后继续实验,将精子培养液BWW加入至空白对照组,比较各组雄激素结合蛋白(ABP)、转铁蛋白(TF)、血清抑制素B(INHB)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)水平及溶酶体相关膜蛋白2(LAMP-2)及微管相关蛋白1轻链3(LC3)水平、各组精子细胞活力。结果与空白对照组相比,采用菟丝子黄酮处理的各组ABP、TF、INHB水平均更高,IL-1β、IL-6水平均更低(P<0.05)。随着菟丝子黄酮浓度升高,ABP、TF、INHB水平呈升高趋势,IL-1β、IL-6水平呈下降趋势(P<0.05)。相比于空白对照组,应用不同浓度菟丝子黄酮处理的各组LC3及LAMP22自噬体蛋白活性更高(P<0.05)。随着菟丝子黄酮浓度不断升高,各组LC3及LAMP22呈升高趋势,至200μmol/L时达到最高值,然后逐渐下降(P<0.05)。与空白对照组相比,采用菟丝子黄酮处理的各组精子细胞活力均更高(P<0.05)。精子细胞活力于菟丝子黄酮浓度达到200μmol/L时达到最高,然后呈下降趋势(P<0.05)。结论菟丝子中黄酮类成分可通过调控睾丸支持细胞线粒体自噬途径的方式影响精子细胞活力。Objective To analyze the effect of flavonoids in Semen Cuscutae on sperm cell viability based on mitochondrial autophagy path-way of sertoli cells.Method The primary sertoli cells were subcultured,then the sertoli cells were treated with different concentrations of Semen Cuscutae flavonoids.After 72 hours of culture,the experiment was continued,and the sperm culture solution BWW was added to the blank control group.The androgen binding protein(ABP),transferrin(TF),serum inhibin B(INHB),interleukin-1β(IL-1β)and interleukin-6(IL-6)were compared among the groups.Result Compared with the blank control group,the levels of ABP,TF and INHB were higher,and the levels of IL-1βand IL-6 were lower(P<0.05).With the increase of flavonoids concentration in Cuscuta chinensis,the levels of ABP,TF and INHB increased,while the levels of IL-1βand IL-6 decreased(P<0.05).Compared with the blank control group,the activity of LC3 and LAMP22 autophagy protein in each group treated with different concentrations of Semen Cuscutae flavonoids was higher(P<0.05).With the increasing concentration of flavonoids in Semen Cuscutae,LC3 and LAMP22 in each group showed an increasing trend,reaching the highest value at 200μmol/L,and then gradually decreased(P<0.05).Compared with the blank control group,the sperm cell via-bility of each group treated with Semen Cuscutae flavonoids was higher(P<0.05).Sperm cell viability reached the highest when the concen-tration of flavonoids in Cuscuta chinensis reached 200μmol/L,and then showed a downward trend(P<0.05).Conclusion Flavonoids in Semen Cuscutae can affect sperm cell viability by regulating the mitochondrial autophagy pathway of sertoli cells.
