摘  要:【目的】为确定新疆阿克苏地区某规模化猪场产房仔猪出现运动失调、抽搐、昏迷、关节肿胀、疼痛和淋巴结化脓的发病原因,从疑似链球菌感染的病死仔猪的脑组织中进行猪链球菌的分离和鉴定。【方法】对该猪场病死仔猪脑组织进行无菌采样及细菌分离鉴定,使用16S rRNA基因的细菌通用引物进行PCR扩增及测序分析,并进行药敏试验、毒力基因和耐药基因检测。【结果】从疑似患病仔猪猪脑组织中分离出1株猪链球菌。该菌株在BHI固体培养基上显现出了针尖般大小、呈现灰白色且略带透明的菌落,在光学显微镜下可观察到菌体成双或呈长短不一的链状排列。通过PCR方法扩增出16S rRNA 1 500 bp的条带;16S rRNA测序结果与猪链球菌参考菌株相似性位于99.64%~100%之间;药敏结果显示,该菌株对青霉素、氨苄西林、大观霉素等抗生素敏感,对链霉素等抗生素中介,对红霉素、庆大霉素、新霉素等抗生素耐药,毒力基因结果显示该菌株携带了purD、mrp,2种毒力基因;耐药基因结果显示该菌株携带了氨基糖苷类aph(3')Ia基因和大环内酯类ermb、mefA基因。【结论】从疑似患病猪猪脑组织中分离出1株猪链球菌,通过毒力基因和耐药基因的检测分析其致病力和耐药性。以期为该猪场提供科学合理的治疗方案。临床建议减少选用氨基糖苷类和大环内酯类药物治疗。为阿克苏及周边地区猪链球菌的防控和耐药机理研究提供了理论依据。[Objective] In order to determine the causes of dyskinesia,convulsion,coma,joint swelling,pain and lymph node abscess in piglets in a large-scale pig farm in Aksu,Xinjiang,Streptococcus suis was isolated and identified from the brain tissue of dead piglets suspected to be infected with streptococcus.[Methods] The brain tissue of dead piglets in this farm was sampled aseptically and the bacteria were isolated and identified.The 16S rRNA gene universal primers were used for PCR amplification and sequencing analysis,and the drug susceptibility test,virulence gene and drug resistance gene were detected.[Results] A strain of Streptococcus suis was isolated from the brain tissue of suspected diseased piglets.On the BHI solid medium,this strain showed pinpoint-sized,grayish white and slightly transparent colonies,which were arranged in pairs or chains of different lengths under the optical microscope.The 1 500 bp band of 16S rRNA was amplified by PCR.16S rRNA sequencing results were 99.64%~100% similar to the reference strains of Streptococcus suis.The results of drug sensitivity showed that the strain was sensitive to antibiotics such as penicillin,ampicillin,and Daguanomycin,was intermediate to antibiotics such as streptomycin,and was resistant to antibiotics such as erythromycin,gentamicin,and neomycin.The results of virulence genes showed that the strain carried two virulence genes,purD and mrp.The results of drug resistance genes showed that the strain carried aminoglycoside aph(3')Ia gene and macrolide ermb and mefA genes.[Conclusion] A strain of Streptococcus suis was isolated from the brain tissue of suspected infected pigs,and its pathogenicity and drug resistance were analyzed through the detection of virulence gene and drug resistance gene.In order to provide a scientific and reasonable treatment plan for the pig farm.It is recommended to reduce the use of aminoglycosides and macrolides for treatment.The study provided a theoretical basis for the prevention and control of Streptococcus suis and the mechanism
