摘  要:为分析临床腹泻发病猪场疫情发生的原因,在开展初生仔猪腹泻样品病毒分离试验中,获得了能够在Marc145细.胞上稳定增殖的病毒,并对该病毒进行宏基因组测序、核酸序列分析、培养特性研究、仔猪回归试验等研究。该病毒的分离株鉴定为猪肠病毒G1型(EV-G1),能在Marc145、ST、Vero等多种细胞上生长,无胰酶依赖性,对氯仿不敏感,适应细胞培养滴度均能达到107.0 TCIDg/mL以上。人工感染3~5日龄仔猪,可导致一过性腹泻,在感染仔猪的脑、心脏、空肠、回肠、盲肠、结肠中均可检测到该病毒。本研究从与猪轮状病毒混合感染的组织样品中分离到1株EV-G病毒,该病毒可导致新生仔猪腹泻,且病毒有良好的细胞适应性,旨在充分了解仔猪感染EV-G病毒的致病性,为EV-G免疫原性研究以及疫苗研发奠定基础。To analyze the causes of outbreaks in swine farms with clinical diarhea incidence.In conducting virus isolation experiments on diarhea samples from newborn piglets,viruses capable of stable proliferation on Marc 145 cells were obtained.The virus was also subjected to macro-genome sequencing.nucleic acid sequence analysis,culture characterization,and piglet regression testing.The isolate of this virus was identified as porcine enterovirus type G1(EV-G1).which could grow on a variety of cells including Marc145,ST,and Vero without trypsin dependence,was insensitive to chloroform,and was adapted to the cell culture titer could reach more than 1070 TCIDs/mL in all cases.Artificial infection of 3 to 5-day--old piglets could lead to transient diarhea,and the virus could be detected in the brain,heart,jejunum,ileum,cecum,and colon of infected piglets.In this study,one strain of EV-_G virus was isolated from tissue samples mixed with porcine rotavirus infection,which could cause diarrhea in newborn piglets,and the virus had good cellular adaptability,The aim was to fully understand the pathogenicity of EV-G virus infection in piglets and to lay the foundation for EV-G immunogenicity studies as well as vaccine development.
