摘  要:胶束作为良好的肿瘤药物载体,不同形态胶束在肿瘤组织的穿透力不同。通过研究在胆固醇/DSPE-m PEG2000/DPPC系统中, DSPE-m PEG2000的含量对盘状和球状胶束结构转变的影响,寻找盘状胶束完全变构为球状胶束的临界点。实验结果表明,随着处方中DSPE-m PEG2000的量增加,脂质体先破裂形成盘状胶束,然后盘状胶束合拢变构为球状胶束,然后出现了单独由DSPE-m PEG2000形成的小粒径聚合物胶束。处方中加入40%胆固醇的情况下, DPPC和DSPE-m PEG2000含量分别为47.1%和12.9%,此组分比例是本实验探索得到的临界点。As a good drug carrier for tumor,micelle of different forms have different penetration in tumor tissue.The effect of DSPE-mPEG2000 content on the structure transition of discoid and globular micelles in the cholesterol/DSPEm-PEG2000/DPPC system was investigated to find the critical point for the complete allosteric formation of discoid micelles into globular micelles.The experimental results showed that as the amount of DSPE-mPEG2000 in the formulation increased,the liposomes first ruptured to form disk-like micelles,and then the disk-like micelles were closed and allosteric to spherical micelles,and then the small particle size polymeric micelles formed by DSPE-mPEG2000 alone appeared.When 40%cholesterol was added,DPPC and dspe-mPEG2000 contents were 47.1%and 12.9%,respectively,which was the critical point explored in this experiment.
