上调miR-224-3p对脓毒症大鼠肺损伤及TLR4/NF-κB通路的影响

摘  要:目的探究上调miR-224-3p对脓毒症大鼠肺损伤及Toll-样受体4(TLR4)/核因子-κB(NF-κB)通路的影响。方法将64只SD大鼠随机均分为假手术组、模型组、miR-224-3p模拟物(mimics)组、mimics-control组。采用盲肠结扎穿孔术建立脓毒症肺损伤大鼠模型。ELISA检测各组大鼠血清炎症因子水平;测量肺组织湿/干质量(W/D)比值评估肺水肿的严重程度;HE染色观察各组肺组织病理变化;qRT-PCR检测各组肺组织miR-224-3p表达水平;Western blotting检测肺组织TLR4/NF-κB通路相关蛋白表达情况。结果模型组、mimics-control组大鼠肺组织病理损伤严重,大量炎症细胞浸润;miR-224-3p mimics组大鼠肺组织病理损伤明显减轻,少量炎症细胞浸润。与假手术组比较,其他各组大鼠肺组织miR-224-3p表达降低,血清炎症因子水平、肺组织W/D比值、TLR4/NF-κB通路相关蛋白表达均升高(P<0.05)。与模型组或mimics-control组比较,miR-224-3p mimics组大鼠肺组织miR-224-3p表达升高,血清炎症因子水平、肺组织W/D比值、TLR4/NF-κB通路相关蛋白表达均降低(P<0.05)。结论上调miR-224-3p可减轻脓毒症大鼠肺损伤,其作用机制可能与抑制TLR4/NF-κB信号通路的活化有关。Aim To investigate the effects of upregulating miR-224-3p on lung injury and Toll-like receptor 4(TLR4)/nuclear factor-κB(NF-κB)pathway in sepsis rats.Methods Sixty-four SD rats were randomly divided into sham surgery group,model group,miR-224-3p mimics group and mimics-control group.The rat model of sepsis-induced lung injury was established by cecal ligation and perforation.The levels of inflammatory factors were detected by ELISA.The wet/dry(W/D)weight ratio of lung tissue was measured to assess the severity of pulmonary edema.The pathological changes in lung tissue of each group was observed by HE staining.The expression of miR-224-3p in lung tissue was detected by qRT-PCR.The expression of TLR4/NF-κB pathway related proteins in lung tissue was detected by Western blotting.Results The pathological damage to the lung tissue of rats in the model group and the mimics-control group was severe,accompanying with a large number of inflammatory cell infiltration.However,the pathological damage to the lung tissue of rats in the miR-224-3p mimics group was significantly reduced,with only a small amount of inflammatory cell infiltration.Compared with the sham surgery group,the expression of miR-224-3p in the lung tissue of rats in other groups were decreased,while the levels of serum inflammatory factors,W/D ratio in lung tissue,and expression of TLR4/NF-κB pathway related proteins were increased(P<0.05).Compared with the model group or mimics-control group,the above indicators of rats were reversed in the miR-224-3p mimics group(P<0.05).Conclusion Upregulation of miR-224-3p can reduce lung injury in sepsis rats,and its mechanism may be related to the inhibition of TLR4/NF-κB signaling pathway activation.
