摘 要:目的:采用高通量测序研究灵芝醇提物对APP/PS1小鼠肠道菌群多样性及组成的影响。方法:APP/PS1小鼠随机分为模型组、灵芝组,每组10只,C57BL/6J小鼠为对照组(n=10),模型组和对照组用灵芝组等体积蒸馏水处理持续6个月,行为学评价后采集各组小鼠新鲜粪便样本,进行微生物群落多样性测序分析,比较各组小鼠肠道菌群组成和结构。结果:灵芝组与模型组小鼠肠道菌群组成存在显著差异。与模型组比较,灵芝组Akkermansia,Enterobacter,Coprobacillus、Clostridium丰度显著增高,差异有统计学意义(P<0.05),Adlercreutzia,Alistipes,Anaerotruncus,Bacillus丰度显著降低,差异有统计学意义(P<0.05)。结论:灵芝醇提物可增加APP/PS1小鼠肠道Akkermansia、Enterobacter、Coprobacillus、Clostridium等有益菌群的丰度,降低肠道Adlercreutzia、Alistipes、Anaerotruncus、Bacillus等有害菌群丰度,影响小鼠肠道菌群组成与比例。Objective To study the effects of Ganoderma lucidum alcohol extract on intestinal flora diversity and composition in APP/PS1 mice using high-throughput sequencing.Methods APP/PS1 mice were randomly divided into model group,Ganoderma lucidum group,and control group(consisting of C57BL/6J mice).Each group contained 10 animals.The model group and control group were administered equal volumes of distilled water,while the Ganoderma lucidum group received the extract.After 6 months of treatment,behavioral evaluations were conducted.Fresh fecal samples were collected from mice in each group,and microbial community diversity sequencing and interactive analysis were performed.The results were analyzed using bioinformatics software to compare the composition and structure of intestinal flora among the groups.Results Significant differences in intestinal flora were observed between the Ganoderma lucidum group and the model group.Compared with the model group,the abundances of Akkermansia,Enterobacter,Coprobacillus,and Clostridium increased significantly(P<0.05),while those of Adlercreutzia,Alistipes,Anaerotruncus,and Bacillus decreased significantly(P<0.05)in the Ganoderma lucidum group.Conclusions The Ganoderma lucidum alcohol extract could increase the abundance of certain beneficial bacteria and decrease the abundance of certain harmful bacteria in the intestinal flora of mice,thereby affecting the composition and proportion of the intestinal flora.
