摘  要:目的探究鱼酱排毒鼻喷雾剂(Yujiang Paidu Nasal Spray,YJNS)对金黄色葡萄球菌(Staphylococcus aureus,S.aureus)的体外抑菌机制。方法采用微量肉汤稀释法检测YJNS对S.aureus的最低抑菌浓度(MIC)及最低杀菌浓度(MBC);绘制细菌生长曲线,初步确定对S.aureus的抑菌活性;采用结晶紫半定量法,测定YJNS对S.aureus生物膜的影响;试剂盒检测S.aureus培养液中碱性磷酸酶(AKP)的含量、酶标仪检测特定波长下检测β-半乳糖苷酶和肽聚糖(PG)的吸光值,评估其对金黄色葡萄球菌细胞壁、细胞膜的影响;电镜下观察YJNS作用前后菌体细胞形态变化。最后通过琼脂糖凝胶电泳检测对S.aureus DNA基因组的影响。结果YJNS对S.aureus的MIC、MBC均为6.25mg/mL。与空白组相比,YJSN作用下的S.aureus生物膜形成量明显降低(P<0.01);S.aureus培养液中AKP、β-半乳糖苷酶含量和菌体内的PG的吸光值均显著增加(P<0.01);扫描电镜下S.aureus细胞形态发生变化,出现皱缩、塌陷、细胞内容物外泄等现象;S.aureus基因组出现断裂。结论鱼酱排毒鼻喷雾剂可通过破环金黄色葡萄球菌细胞壁、细胞膜,导致菌体内容物外泄,进而影响基因组,最终发挥抑菌作用。该研究为鱼酱排毒鼻喷雾剂用于急性鼻窦炎的治疗,提供理论基础。Objective To investigate the in vitro bacteriostatic mechanism of Yujiang Paidu Nasal Spray(YJNS)against Staphylococcus aureus(S.aureus).Methods Determination of minimum inhibitory Concentration(MIC)and minimum bactericidal Concentration(MBC)of YJNS against S.aureus by microbroth dilution method;The bacterial growth curve was drawn to determine the bacteriostatic activity against S.aureus;The effect of YJNS on S.aureus biofilm was determined by crystal violet semi-quantitative method;The content of alkaline phosphatase(AKP)in S.aureus culture medium was detected by the kit,and the absorbance values ofβ-galactosidase and peptidoglycan(PG)were detected by the enzyme label under specific wavelength,and their effects on the cell wall and membrane of S.aureus were evaluated.The morphological changes of somatic cells before and after YJNS treatment were observed under scanning electron microscope.Finally,the effect of agarose gel electrophoresis on S.aureus DNA genome was detected.Results The MIC and MBC of YJNS against S.aureus were 6.25mg/mL.Compared with blank group,the amount of S.aureus biofilm formation under YJSN was significantly decreased(P<0.01);The contents of AKP andβ-galactosidase in S.aureus culture medium and the light absorption value of PG in vivo were significantly increased(P<0.01);Under scanning electron microscope,the morphology of S.aureus cells was changed,and the cell contents were wrinkled,collapsed,and leaked;The S.aureus genome is broken.Conclusion Yujiang Paidu Nasal Spray can break the cell wall and membrane of Staphylococcus aureus,resulting in the release of bacterial contents,and then affect the genome,and finally play an antibacterial role.This study provides a theoretical basis for the use of Yujiang Paidu Nasal Spray in the treatment of acute sinusitis.
