摘  要:目的探究外泌体环状RNA诱导HK-2细胞凋亡在感染性休克相关性急性肾损伤的作用机制。方法分为感染性休克相关性急性肾损伤患者组和健康人对照组,采集血标本。检测外泌体标记蛋白,鉴定出两组外泌体,对外泌体环状RNA进行实时PCR分析。对两组的HK-2细胞进行凋亡检测。结果10例感染性休克AKI入院患者,10例健康人对照。鉴定表面外泌体标记蛋白包括HSP70和TSG101。两组均鉴定出外泌体。与对照组相比,感染性休克AKI组circ-Ttc3(P<0.05)、circ-Fryl(P<0.01)表达明显下调。感染性休克AKI外泌体可抑制HK-2细胞活性,促进细胞凋亡。结论感染性休克AKI可诱导HK-2细胞凋亡,其分子机制可能与circ-Ttc3和circ-Fryl这两种环状RNA有关。Objective To investigate the mechanism of exosomal circular RNA induced HK-2 cell apoptosis in septic shock related acute kidney injury.Methods Blood samples were collected from patients with acute kidney injury associ-ated with septic shock and healthy controls.Two groups of exosomes were identified by detecting exosome labeled pro-teins,and the exosome circular RNA was analyzed by real-time PCR.The apoptosis of HK-2 cells in both groups were detected.Results There were 10 hospitalized patients with septic shock AKI and 1o healthy controls.Surface exosome labeled proteins were identified,including HSP70 and TSG101.Exosomes were identified in both groups.Compared with the control group,the expressions of circ-Ttc3(P<0.05)and circ-Fryl(P<0.01)in AKI group were significantly down-regulated.AKI exosomes in septic shock can inhibit the activity of HK-2 cells and promote cell apoptosis.Conclu-sion AKI can induce the apoptosis of HK-2 cells,and the molecular mechanism may be related to circ-Ttc3 and circ-Fryl.
