摘 要:目的:建立不同产地酒制多花黄精中糖类成分的定性与定量方法。方法:以11个产地的酒制多花黄精为研究样品,采用分子排阻/配体交换色谱-示差折光检测法(MEC/LEC-RI)分析酒制多花黄精糖类成分分子量,构建MEC-RI指纹图谱,测定D-葡萄糖、D-果糖含量,采用硫酸-蒽酮-紫外光谱法测定酒制多花黄精中黄精多糖含量,对糖类成分定性与定量结果进行关联性分析。结果:酒制多花黄精水浸液中主要含有分子量低于1 000 Da的低聚多糖,从MEC-RI指纹图谱中指认了5个共有峰,分子量分别约为220k、160k、100k、14.5k、0.95k Da,11个产地样品相似度均大于0.9。酒制多花黄精中D-葡萄糖、D-果糖含量在58.771~116.684、143.087~162.054 mg/g范围之间,黄精多糖含量在42.769~76.065 mg/g范围之间。结论:建立了不同产地酒制多花黄精中糖类成分的定性与定量方法,比较了不同产地酒制多花黄精中糖类成分的差异,可为多花黄精质量标准研究、资源分布及产品开发提供科学依据。【Objective】To establish the qualitative and quantitative methods of saccharides in the Polygonatum cyrtonema from differ-ent producing areas.【Method】Polygonatum cyrtonema from 11 producing areas as the research sample,the molecular weight of sac-charides was analyzed by Molecular Exclusion Chromatography-Differential Refraction(MEC-RI),the MEC-RI fingerprint chromatography was constructed by MEC-RI,the content of D-glucose,D-fructose was measured by Ligand Exchange Chromatography-Differential Refraction(LEC-RI),and the content of polysaccharide was measured by Sulfuric Acid-Phenol-Ultraviolet Spectrum(UV),correlation analysis was carried out between qualitative and quantitative results of carbohydrate components.【Result】The oligopolysac-charide with molecular weight lower than 1000Da were mainly contained in the aqueous extract of Polygonatum cyrtonema,the molecular weights of 5 common peaks were about 220k,160k,100k,14.5k and 0.95k Da respectively were identified from MEC-RI fingerprints,the similarity of samples from 11 producing areas was all greater than 0.9,the contents of D-glucose and D-fructose in Polygonatum cyrtonema were between 58.771—116.684,143.087—162.054 mg/g,the total polysaccharide content was in the range of 42.769—76.065 mg/g.【Conclusion】The qualitative and quantitative methods of polysaccharide in Polygonatum cyrtonema from different producing areas were established,and the differences of polysaccharide in Polygonatum cyrtonema from different areas were compared,which can provide scientific basis for quality standard research,resource distribution and product development of Polygonatum cyrtonema.
