摘 要:[目的]提高多粘类芽孢杆菌产孢和产IAA能力以及对花生的促生效果。[方法]选择LB、YD和YDC3种发酵培养基进行条件优化,并通过盆栽试验测定多粘类芽孢杆菌不同发酵液拌种对花生发芽率、根长和鲜重的影响。[结果]多粘类芽孢杆菌在LB、YD和YDC3种发酵培养基中产孢和产IAA能力存在显著差异。YDC培养基(马铃薯200 g、玉米粉25 g、黄豆粉25 g、葡萄糖5 g、茶饼粉10 g、水1000 mL)更有利于芽孢和IAA的产生,芽孢数达57.62×10~8 CFU/mL,是LB、YD培养基中芽孢数量的9.29倍和5.85倍;IAA含量为3.08μg/mL,较LB和YD发酵培养基分别增加了52.48%和22.71%;盆栽试验显示,多粘类芽孢杆菌各发酵液对花生的发芽率、根长和鲜重均有一定的促生效果,能够有效促进花生种子的萌发和幼苗的生长,且YDC发酵液促生效果优于其他处理;此外相关性分析发现,芽孢数、IAA含量与花生发芽率、根长等呈显著正相关。[结论]多粘类芽孢杆菌能够有效促进花生种子的萌发和幼苗生长,且该菌在YDC发酵培养基更有利于多粘类芽孢杆菌芽孢的形成和IAA的产生。[Objective]To improve the ability of Bacillus polymyxoides to produce spores and IAA and to promote the growth of peanut.[Method]LB,YD and YDC fermentation medium were selected to optimize the conditions,and the effects of different fermentation solution of Bacillus polymyces on peanut germination rate,root length and fresh weight were determined by pot experiment.[Result]There were significant differences in the ability of P.polymyxa to produce spores and IAA in LB,YD and YDC fermentation medium.YDC medium(potato 200 g,corn flour 25 g,soybean flour 25 g,glucose 5 g,tea cake powder 10 g,water 1000 mL)was more conducive to the production of spores and IAA,the number of spores reached 57.62×10~8 CFU/mL,which was 9.29 times and 5.85 times of that in LB and YD medium.IAA content was 3.08μg/mL,which was increased by 52.48%and 22.71%compared with LB and YD medium.The pot experiment showed that each fermentation broth of P.polymyxa had certain promoting effect on the germination rate,root length and fresh weight of peanut,which could effectively promote the germination of peanut seeds and the growth of seedlings,and the growth promotion effect of YDC fermentation broth was better than other treatments.In addition,the correlation analysis showed that spore number and IAA content were significantly positively correlated with peanut germination rate and root length.[Conclusion]Bacillus polymyxis can effectively promote peanut seed germination and seedling growth,and it is more conducive to the formation of P.polymyxa spores and the production of IAA in YDC fermentation medium.
