摘  要:目的探讨A2A受体在心脏衰老中的作用。方法不同喂养方法持续喂养16周龄成年小鼠及56周龄老年小鼠30周来分别建立成年小鼠及老年小鼠模型。将小鼠分成四组:成年小鼠标准喂养组、成年小鼠标准喂养加A2A受体抑制剂[1 mg/(kg·d)]组,老年小鼠标准喂养组、老年小鼠标准喂养加A2A受体抑制剂[1 mg/(kg·d)]组,观察A2A受体抑制剂对成年小鼠和老年小鼠心肌重构、心脏功能、心脏间质纤维化的影响。结果与成年小鼠标准喂养组相比,老年小鼠标准喂养组心肌组织A2A受体表达升高(t=4.697,P<0.05)。与老年小鼠标准喂养组相比,补充A2A受体抑制剂喂养减少老年小鼠心肌细胞肥大,缩小老年小鼠心肌细胞横切面面积(t=3.710,P<0.05),改善左心室质量、左心室后壁舒张末期厚度、左心室舒张末期内径、左心室舒张末期体积(t=2.327、3.411、3.898、3.844,P<0.05),提高心脏收缩功能射血分数、缩短分数值(t=7.941、5.042,P<0.05),抑制心脏间质纤维化(t=3.147,P<0.05)。结论A2A受体与心脏衰老密切相关,而抑制A2A受体对老年心脏具有保护作用。Objective To investigate the role of A2A receptor in cardiac aging process.Methods Adult and elderly mouse models were established respectively by continuously feeding 16-week-old adult mice and 56-week-old elderly mice for 30 weeks using different feeding methods.The mice were randomly divided into four groups:adult standard diet group,adult standard diet plus A2A receptor inhibitors[1 mg/(kg·d)]group,aged standard diet group,and aged standard diet plus A2A receptor inhibitors[1 mg/(kg·d)]group.The effects of the A2A receptor inhibitor on myocardial hypertrophy,cardiac function,and cardiac interstitial fibrosis in adult and aged mice were observed.Results Compared with adult standard diet group,the expression of A2A receptor in myocardial tissue increased in the aged standard diet group(t=4.697,P<0.05).Compared with the aged standard diet group,the supplementation of the A2A receptor inhibitor reduced myocardial cell hypertrophy in aged mice,decreased the cross-sectional area of myocardial cells in aged mice(t=3.710,P<0.05),improved left ventricular mass,left ventricular posterior wall thickness,left ventricular end diastolic diameter,and left ventricular end diastolic volume(t=2.327,3.411,3.898,3.844;P<0.05),increased cardiac systolic function ejection fraction,fractional shortening(t=7.941,5.042;P<0.05),and inhibited cardiac interstitial fibrosis(t=3.147,P<0.05).Conclusion The A2A receptor is closely related to cardiac aging,and inhibiting the A2A receptor has a protective effect on the aging heart.
